Description	Lanreotide, a long-acting somatostatin analog, demonstrated potent preclinical activity by binding selectively to somatostatin receptor subtypes sst2 and sst5 with high affinity (IC₅₀ ~0.5–1.0 nM). In rodent models, lanreotide inhibited growth hormone release by up to 80% and reduced insulin and glucagon secretion dose-dependently. In rat pancreatic tumor models, lanreotide significantly suppressed tumor growth and hormone secretion, with up to 60% reduction in tumor volume. Additionally, in xenograft models of neuroendocrine tumors, lanreotide reduced proliferation markers and prolonged survival, supporting its development for acromegaly and hormone-dependent tumors.

Basic Information

Generic Name: Lanreotide acetate

Drug Class: Somatostatin analogs

Mechanism of Action: It binds to somatostatin receptors on target cells, inhibiting the secretion of various hormones. This includes growth hormone (GH) and insulin-like growth factor-1 (IGF-1) in acromegaly, as well as vasoactive intestinal peptide (VIP), serotonin, and other hormones in neuroendocrine tumors.

Main Indications

Acromegaly: Used to control symptoms and reduce levels of growth hormone and insulin-like growth factor-1 in patients with acromegaly who are unresponsive to surgery or radiation therapy, or as a first-line treatment.

Neuroendocrine Tumors: Indicated for the symptomatic treatment of unresectable, well-differentiated gastroenteropancreatic neuroendocrine tumors (GEP-NETs) and carcinoid tumors, relieving symptoms such as flushing and diarrhea.

Thyrotropin-Secreting Pituitary Adenomas (TSHomas): Used to reduce thyroid-stimulating hormone (TSH) levels and control thyroid function in patients with this type of adenoma.

Administration and Dosage

Route of Administration: Subcutaneous injection or intramuscular injection of long-acting depot formulations.

Dosage Characteristics: The dosage is individualized based on the patient's disease type, severity, and treatment response. Long-acting formulations can be administered once every 4 weeks or 8 weeks, improving patient compliance.

Adverse Reactions
