    Description

    In vitro, Tenofovir (most often studied as its prodrugs TDF or TAF) inhibits HIV-1 and HBV replication with EC₅₀ values typically 0.005–0.5 μM across various cell systems. It lowers viral DNA synthesis by >90% at submicromolar doses. Against HBV-infected hepatocyte models, Tenofovir reduces extracellular HBV DNA levels by 2–4 logs. In vivo, Tenofovir demonstrates strong antiviral efficacy in animal models: in HIV-infected humanized mice, TDF or TAF dosing (e.g., 10–25 mg/kg) reduces plasma viral load by 2–3 logs. In woodchuck hepatitis virus–infected woodchucks, Tenofovir decreased viral DNA by 3–5 logs, with improvements in liver histology. It is generally well tolerated in short-term animal studies.
