PNC-27 is a synthetic anticancer peptide derived from the tumor suppressor protein p53. With its unique molecular design and mechanism of action, it has shown great potential in targeted therapy research for various cancers while minimizing damage to normal cells.

Developed by Dr. Joseph M. Dahabieh and his team at the State University of New York Downstate Medical Center in the early 21st century, the peptide was initially designed to mimic the MDM-2 binding domain in p53, achieving targeted anticancer effects.

Precise Targeting of Cancer Cells:

HDM-2 protein is a key regulatory factor that inhibits p53 function. It is highly overexpressed on the cell membranes of various cancer cells, while its expression level is extremely low or absent in normal cells. PNC-27 can bind to HDM-2 on cancer cell membranes with high affinity. This targeting ability is the core reason for its distinction between cancer cells and normal cells, as well as an important factor contributing to its low toxicity.

Synergistically Enhancing the Efficacy of Other Drugs:

Studies have found that PNC-27 can synergize with various anticancer drugs such as rapamycin, methotrexate, and paclitaxel. It can significantly reduce the half-maximal inhibitory concentration required for these drugs to kill cancer cells and enhance their anticancer effects. Therefore, it is expected to be used as an adjuvant therapeutic drug in combined anticancer regimens.
