Betulinic acid

Betulinic acid (CAS Number: 472-15-1) is a naturally occurring lupane-type pentacyclic triterpenoid derived mainly from the bark of white birch trees (Betula species). It functions as a highly selective anti-cancer, anti-inflammatory, and anti-retroviral research compound. Unlike many standard chemotherapeutics, it exhibits tumor-specific cytotoxicity, meaning it destroys cancer cells while leaving healthy non-malignant tissues largely unaffected. 

• Selective Induction of Mitochondrial Apoptosis: Its most celebrated function is triggering programmed cell death exclusively in tumor cells. It directly permeabilizes the outer mitochondrial membrane, inducing the release of cytochrome c into the cytosol. This activates the apoptotic caspase cascade (specifically caspases-3, -8, and -9) via a mechanism that bypasses p53 and CD95 pathway resistance.
• Dual Anti-Inflammatory and Immunomodulatory Potential: It blocks the NF-\(\kappa\text{B}\) signaling pathway by preventing the phosphorylation and degradation of I\(\kappa\text{B}\). This stops the expression of inflammatory enzymes like COX-2 and down-regulates a host of pro-inflammatory cytokines, including TNF-\(\alpha \) and IL-1\(\beta \). Concurrently, it increases protective IL-10 production.
• Antiretroviral and Anti-HIV Mechanisms: It acts as a powerful structural lead for HIV-1 maturation inhibitors. It prevents the proper cleavage of the viral Gag precursor protein during replication, resulting in the assembly of non-infectious, immature viral particles.
• Metabolic Syndrome and Weight Homeostasis: Betulinic acid serves as a TGR5 receptor agonist. In metabolic models, it actively regulates glucose, adipose tissue, and carbohydrate pathways. This suppresses hyperlipidemia (high blood fats), enhances insulin sensitivity, and assists in controlling body weight.
• Autophagy Enhancement and Neuroprotection: It readily crosses the blood-brain barrier, where it counters neuroinflammation and oxidative stress. In neurological injury models, it activates the AMPK/mTOR/TFEB pathway to boost cytoprotective autophagy. This process efficiently clears toxic cell accumulations and suppresses cellular pyroptosis.
• Broad Antimicrobial and Antimalarial Action: It shows direct therapeutic efficacy against severe protozoal and bacterial infections. It disrupts topoisomerase activity and targets internal pathogen structures to curb Plasmodium falciparum (malaria) and various bacterial strains.
