Dihexa (CAS 1401708-83-5), also known as PNB-0408 or N-hexanoic-Tyr-Ile-(6)-amino hexanoic amide, is a novel, potent, orally active, brain-penetrating oligopeptide drug derived from angiotensin IV . It binds with exceptionally high affinity to hepatocyte growth factor (HGF) with a Kd value of 65 pM and potentiates its activity at the c-Met receptor .

Mechanism of Action:

Dihexa functions as an activator of the HGF/c-Met system. It binds to HGF with high affinity and, in the presence of subthreshold concentrations of HGF, induces c-Met phosphorylation and augments HGF-dependent cell scattering . Both Dihexa and its parent compound Norleucine 1-AngIV induce hippocampal spinogenesis and synaptogenesis, similar to the effects of HGF itself . At 1 μM, Dihexa effectively inhibits HGF dimerization .
