Tofacitinib (CAS 477600-75-2), also known as Tasocitinib or CP-690550, is an orally bioavailable Janus kinase (JAK) modulator that selectively targets JAK3 with an IC₅₀ of 1 nM, and shows lower activity against JAK2 and JAK1 (IC₅₀ = 20 and 112 nM, respectively) . It appears as a white to off-white solid powder with the molecular formula C₁₆H₂₀N₆O and a molecular weight of 312.37 g/mol .

With a purity of ≥98.0% by HPLC, Tofacitinib is a well-established pharmaceutical compound first approved by the FDA in 2012 (brand name Xeljanz) for the treatment of moderate to severe rheumatoid arthritis. It has since received additional approvals for psoriatic arthritis and ulcerative colitis
