Methyl lysergate is a tryptamine derivative with 5-HT1 receptor agonist activity. It is widely applied in the research and preparation of biomedical intermediates.

Under vigorous stirring, a solution of diazomethane in diethyl ether (0.5 M, 75.0 mmol, 150 mL) was added to a suspension of (6aR,9R)-7-methyl-4,6,6a,7,8,9-hexahydroindolo[4,3-fg]quinoline-9-carboxylic acid (2.01 g, 7.5 mmol) in anhydrous methanol (300 mL). The mixture was stirred until the solution became clear. The resulting mixture was concentrated under vacuum, and the residue was dispersed in dichloromethane (100 mL). Insoluble solids were removed by filtration, and the filter cake was washed with dichloromethane (3 × 30 mL). The combined filtrate was concentrated under vacuum to obtain the target product, (6aR,9R)-7-methyl-4,6,6a,7,8,9-hexahydroindolo[4,3-fg]quinoline-9-carboxylic acid methyl ester (methyl lysergate), as an off-white foamy solid.
