2. Chemical Properties

The product appears as white powder recrystallized from DMF-water solution. The hemihydrate has a melting point of 288-290 °C (decomposition).

(-)-configuration: Colorless needle crystals obtained from DMF, melting point 305-306 °C (decomposition), specific rotation [α]D20 -116.7° (C=1.0, DMF).

(+)-configuration: Colorless needle crystals obtained from DMF, melting point 305-306 °C (decomposition), specific rotation [α]D20 +116.9° (C=1.0, DMF).

3. Product Application

Rebamipide is a fine chemical intermediate, widely used in fine chemical synthesis and chemical raw material research.

4. Synthesis Method

4.1 Preparation of Racemic Rebamipide

Using methyl 2(1H)-quinolinone-4-carboxylate as the raw material, it is reduced with lithium borohydride to obtain 2(1H)-quinolin-4-ylmethanol, and then reacted with hydrobromic acid to form 4-(bromomethyl)-2(1H)-quinolinone.

Diethyl acetamidomalonate (12.0 g, 55 mmol) and sodium metal (1.5 g, 65 mmol) were dissolved in 150 mL of anhydrous ethanol. The solution was heated to boiling, and 4-(bromomethyl)-2(1H)-quinolinone (12.0 g, 50 mmol) was added under stirring. The mixture was refluxed for 2 hours, concentrated under reduced pressure, and the residue was poured into water. The precipitate was collected by filtration and recrystallized from ethanol to obtain colorless prismatic crystals with a yield of 69% and melting point of 224-226 °C (decomposition).

The obtained compound (5.0 g, 13 mmol) was suspended in 150 mL of 20% hydrochloric acid solution and refluxed for 9 hours. The solution was concentrated to dryness under reduced pressure, and the residue was recrystallized from ethanol-water solvent to obtain colorless prismatic crystals with a yield of 89% and melting point of 220-225 °C (decomposition).

The intermediate product (2.7 g, 10 mmol) and potassium carbonate (4.2 g, 30 mmol) were dissolved in a mixed solvent of 100 mL acetone and 20 mL water. Under ice bath cooling and continuous stirring, 4-chlorobenzoyl chloride (2.1 g, 12 mmol) was dropwise added, and the system was stirred for another 2 hours. Acetone was removed by vacuum distillation, and the residual liquid was acidified with dilute hydrochloric acid. The precipitate was filtered and recrystallized from methanol-water to obtain white powdery rebamipide with a yield of 49% and melting point of 288-290 °C (decomposition).

4.2 Preparation of (-)-Configuration Product

Racemic rebamipide (21.6 g) was suspended in 1100 mL methanol, and 23 g brucine was added. The clear solution was stirred at room temperature for 4 hours and cooled overnight. The precipitate was collected by filtration, washed with cold methanol, and recrystallized four times to obtain pure salt product. The product was suspended in methanol, added with 10% hydrochloric acid, filtered and washed with water, then recrystallized three times with DMF-water to obtain colorless needle-shaped (-)-configuration rebamipide, with a yield of 14.8% and melting point of 305-306 °C.

4.3 Preparation of (+)-Configuration Product

The solid recovered from the mother liquor of (-)-configuration preparation was dissolved in methanol and treated with 10% hydrochloric acid. The precipitate was filtered and washed with water, then suspended in methanol with brucine added. After stirring and filtration, concentrated hydrochloric acid was added to the filtrate. The precipitate was collected and recrystallized four times with DMF-water to obtain colorless needle-shaped (+)-configuration rebamipide, with a yield of 27.8% and melting point of 305-306 °C (decomposition).
