Brief Introduction
Sodium 8-(2-hydroxybenzamido)octanoate, abbreviated as SNAC, is an absorption enhancer for bicarbonate compounds. It can be applied in the relief of gastrointestinal discomfort, especially for gastrointestinal conditions triggered by insufficient absorption of bicarbonate compounds.
Biological Activity
Salcaprozate Sodium (SNAC) is an oral absorption booster, which can potentially serve as a delivery carrier for oral heparin and oral insulin.
Salcaprozate Sodium can boost lipophilicity via non-covalent macromolecular complexation, thus promoting passive transcellular permeation across small intestinal epithelial cells.
In Vitro Research
SNAC (12.5–400 μg/mL; 24 h) shows no toxic effect on Caco-2 cells. Cell viability remains above 90% at a SNAC concentration of 200 μg/mL.
Compared with the Papp value of SA solution, SNAC at concentrations of 50 μg/mL and 200 μg/mL raises the apparent permeability coefficient (Papp) of RA and SA-B by 2.14 times and 3.68 times respectively.
In Vivo Research
SNAC can improve the oral absorption of both R1 and SA and elevate bioavailability in rat models.
When administered orally via gavage at a daily dose of 2000 mg/kg for 13 weeks, mortality was only observed at this dosage group, with a 20% mortality rate in male rats and a 50% mortality rate in female rats; no clear cause of death was identified.
No mortality occurred in Wistar rats receiving daily oral gavage doses ranging from 100 mg/kg to 1000 mg/kg over a 13-week test period.
