Dacomitinib (CAS 1110813-31-4) is an orally available, irreversible pan-ErbB tyrosine kinase inhibitor with the molecular formula C₂₄H₂₅ClFN₅O₂ and a molecular weight of 469.94 g/mol . It is a second-generation EGFR-TKI developed by Pfizer that irreversibly inhibits EGFR (HER1), HER2, and HER4 . The compound features a quinazoline core bearing a 3-chloro-4-fluoroanilino group at the 4-position, a methoxy group at the 7-position, and a 4-(piperidin-1-yl)but-2-enamide side chain at the 6-position .
