Erdafitinib is an orally bioavailable, functionally selective small-molecule inhibitor of fibroblast growth factor receptors 1-4 (FGFR1-4). It inhibits FGFR tyrosine kinase activity (IC50 1.2-5.7 nM), blocking downstream RAS-MAPK/ERK and PI3K-AKT signaling that drives proliferation and survival of tumor cells harboring activating FGFR alterations (mutations, amplifications, or fusions). Rapid lysosomal uptake contributes to prolonged inhibition of FGFR signaling.
