4-Methyl-pyridine-2-carbaldehyde (433 mg, 71. [5percent)] was obtained from 0.25 M 4-methyl- [2-PYRIDINYLMAGNESIUM] bromide of THF solution (20 mL, 5 mmol) with DMF (0.773 [ML, ] [10 MMOL)] at room temperature under argon.Reference Example 55 (b) In a manner similar to that described for the preparation of 4-cyano-2-pyridinecarboxaldehyde, 2-hydroxymethyl-4-methylpyridine (2.60 g, 21.14 mmol) provided 2.0 g (78%) of the 4-methyl-2-pyridinecarboxaldehyde. TLC (silica gel GF): Rf =0.23 acetone-methylene-hexane (0.5:1.5:8.0). 1 H NMR (CDCl3, TMS):delta 10.09 (s, 1H), 8.67 (d, 1H, J=4.93 Hz), 7.81 (s, 1H):delta 7.38 (d, 1H, J=3.78 Hz), 2.48 (s, 3H).General procedure: The equimolar amount 6-(1H-indol-3-yl)-4, 5?-bipyrimidine-2-amine and substituted aldehydes were mixed in a round bottom flask containing 50 mL of ethanol, some droplets of glacial acetic acid were added to the mixture, and it was set on refluxing for 18 h. Solid product was observed on the completion of reaction that was filtered and dried under vacuum and recrystallized using proper solvent [50].
