3-Chloro-2-fluorobenzonitrile Use and Manufacturing

Take dried 2L flask was refluxed for four devices, under nitrogen, into sulfolane 1200g, 280g of potassium fluoride, tetramethylammonium chloride 50g, 2, 3- dichlorobenzonitrile 688g, open stirring slowly raised to 210 reaction was incubated 8h, the control sample, the end of the reaction raw material ≤1percent.The reaction solution was added to a short column distillation, to give 2-fluoro-3-chlorobenzonitrile 560g, content ≥99percent, yield 90percent.General procedure: An oven-dried round bottomed flask (50 mL capacity) equipped with a magnetic stir bar was evacuated and purged with argon. 2-Aminopyridine (1 mmol, 1 euiv), 2-fluorobenzonitrile (1.2 mmol, 1.2 euiv), KtOBu (4 mmol, 4 euiv) and DMSO (5 mL) were added successivelyat room temperature and the reaction mixture was allowed to stir at room temperature for 12 h. Water (10 mL) was added to the reaction mixture and the organic layer was extracted with ethyl acetate (2×20 mL). The combined organic phases were washed with brine and dried over sodium sulfate. The solvent was removed and the residue was purified by column chromatography on silica gel using hexanes/ethyl acetate as an eluent.General procedure: An oven-dried round bottomed flask (50 mL capacity) equipped with a magnetic stir bar was evacuated and purged with argon.
