4-Amino-3-methylpyridine (3.00 g) was dissolved with icecooling in concentrated sulfuric acid (36 mL). Then, fuming nitric acid (4, 72 g) was added dropwise. After stirring at room temperature for 1 h, the solution was heated at 60 C for 14 hours. After cooling to ambient temperature, the reaction mixture was poured on ice and the resulting solution was adjusted to pH 13 by addition of solid KOH. The precipitate was filtered off, washed with water and dried. Yield: 1. 198 g (31. 3percent) 4-amino-3- methyl-5-nitropyridine.Amixture of 4-Amino-3-methyl-5-nitropyridine (1.5 g, 9.79 mmol)Dissolved in 20 ml of 95% ethanol, added stannous chloride(5.57 g, 29.38 mmol).The reaction solution was warmed to reflux and stirred at this temperature for 2-3 hours.Pour the reaction solution into 100 ml of 2N sodium hydroxide solution.Extracted with ethyl acetate (30 mL×3), and the extract was washed with brine.Dry over anhydrous sodium sulfate and concentrate to give a crude material.Crude silica gel column chromatography(petroleum ether / ethyl acetate = 4:1)3, 4-diamino-5-methylpyridine(517 mg, yield 42.9%).
