Mice deficient in uricase develop uric acid nephropathy and diabetes insipidus which can be mitigated with recombinant uricase treatment.


Urate Oxidase Recombinant produced in E.Coli is a tetrameric, non-glycosylated polypeptide chain containing 302 amino acids, having a molecular formula of C1523H2383N417O462S7 and a molecular mass of 34,247 Dalton.The cDNA coding for urate-oxidase was cloned from a strain of Aspergillus flavus . The monomer protein has no intra- or inter-disulfide bridges.

Using yeast as raw material, the sodium phosphate is extracted and then graded with ammonium sulfate, and then the product is obtained by dialysis, column chromatography and ammonium sulfate recrystallization.

Uses

In the determination of serum and urine uric acid.


Uricase is very important for the determination of uric acid in biological fluids. Uricase is an important medical enzyme which can be used to determine urate in clinical analysis. The enzyme Urate oxidase (UO) catalyzes the oxidation of uric acid to 5-hydroxyisourate.


Uricase fromArthrobacter globiformishas been used for coating the electrodes in - biosensor fabrication for the detection of uric acid in urine samples. It has also been used for analysing the effect of Zn, Cu, Mg, and Mn ions on uricase.

Urate oxidase: ACTIVE|XU - indicates a substance exempt from reporting under the Chemical Data Reporting Rule, (40 CFR 711).
