Solutions of cephalothin may darken, especially if stored at room temperature. A slight discoloration does not affect potency. Solutions stored under refrigeration may precipitate, but the precipitate can be redissolved by warming to room temperature with constant agitation.

Methods of Manufacturing

Using fermented cephalosporin C as the raw material, 7-amino cephalosporanic acid (7-ACA) is condensed with thiophene acetyl chloride by protective hydrolysis. The resulting product is extracted with butyl acetate, activated carbon is decolorized, and finally methanol sodium acetate is added Salt formation, crystallization and cefotaxime sodium.

Uses

Cephalothin is a first-generation cephalosporin antibiotic used to study the mechanism of liposome encapsulated antibiotics,strategies for co-opting β-lactamases of Gram-negative bacteria for treatment of antibiotics,and for immunology studies in relation to antibiotics.It is used to study the effect of expression, binding and inhibition of penicillin-binding proteins especially PBP6 on bacterial cell wall mucopeptide synthesis.

Drug Function and Efficacy

First-generation cephalosporin with a broad antibacterial spectrum and strong activity against Gram-positive bacteria; sensitive to penicillinase-producing and non-producing Staphylococcus aureus, coagulase-negative Staphylococci, Streptococcus pyogenes, Streptococcus pneumoniae, group B hemolytic Streptococcus, viridans streptococci, Staphylococcus epidermidis, Corynebacterium diphtheriae, and Bacillus anthracis; highly sensitive to Haemophilus influenzae, Neisseria meningitidis, Moraxella catarrhalis, and Neisseria gonorrhoeae; moderately sensitive to some strains of Escherichia coli, Klebsiella, Salmonella, Shigella, and Proteus; sensitive to Gram-positive anaerobic bacteria, resistant to Bacteroides fragilis; inhibits bacterial cell wall synthesis.
