Doxifluridine Capsules

This product is one of the new fluorinated pyrimidine series drugs. As a prodrug of fluorouracil (5-FU), it is converted into free 5-FU by pyrimidine nucleoside phosphorylase in the body after taking it, thereby exerting its anti-tumor effect. Animal tumor experiments and clinical tests have proved that the content and activity of pyrimidine nucleoside phosphorylase in tumors are significantly higher than those in normal tissues. Therefore, the conversion rate of 5-FU in tumor tissues of this product is also significantly higher than that of normal tissues and organs, in order of cervical cancer, bladder cancer, breast cancer, colorectal cancer and gastric cancer. In particular, the concentration of 5-FU in colorectal, gastric and breast cancer tumor tissues is significantly higher than that in normal tissues and plasma, so it can selectively kill tumor cells while causing less damage to normal tissues. It affects DNA and RNA synthesis respectively, mainly acting on the S phase, and also affects cells in other proliferation phases.

Deoxyfluridine. Chemical name: 5'-deoxy-5-fluorouridine, molecular formula: C9H11FN2O5, molecular weight: 246.20.

This product is a hard capsule, and the contents are white or off-white powder.

This product is used to treat breast cancer, gastric cancer, colorectal cancer, nasopharyngeal cancer, and cervical cancer.

Oral administration: 800-1200 mg per day, divided into 3-4 doses, taken after meals. The dosage can be increased or decreased according to age and symptoms. A course of treatment is 6-8 weeks.

1. Bone marrow suppression mainly includes leukopenia and thrombocytopenia, and occasionally pancytopenia; 2. Digestive system: nausea, vomiting, loss of appetite, abdominal pain, bloating, constipation, stomatitis, etc., occasionally gastric ulcer, glossitis, etc.; 3. Mental and nervous system: orientation or hearing impairment, occasionally walking or sensory impairment, extrapyramidal symptoms, paralysis, urinary incontinence, etc. Occasionally, abnormal sense of smell; 4. Abnormal electrocardiogram, hair loss, pigmentation, urticaria, etc.

Patients who are allergic to this product or are receiving the antiviral drug Sorivudine are contraindicated.

Precautions for children: The safety for premature infants, newborns, infants or children has not been determined (no experience). If it is necessary to use, please be cautious. Precautions for pregnancy and lactation: 1. According to animal experiments (mice), this product has teratogenic effects. It is best not to use it for pregnant women. 2. According to animal experiments (mice), the drug will enter breast milk, so breastfeeding should be stopped when taking the drug during lactation. Precautions for the elderly: It should be used with caution in elderly patients and patients with low physiological functions.

When the antiviral drug Sorivudine is used in combination with this product or fluorouracil, it can hinder the metabolism of the latter, leading to increased concentrations in the blood, significant bone marrow suppression, and severe blood toxicity that can lead to death, so their combined use is strictly prohibited.
