Azasetron Hydrochloride Injection
Function and Efficacy
Pharmacological action: Azasetron hydrochloride is a selective 5-HT3 receptor antagonist, which has a significant inhibitory effect on nausea and vomiting caused by anticancer drugs such as cisplatin. Animal studies have shown that the affinity of azasetron hydrochloride for the 5-HT3 receptor in the rat cerebral cortex is about 410 times stronger than that of metoclopramide, twice that of ondansetron, and basically the same as granisetron. Toxicological studies: Repeated dose toxicity: This product was administered intravenously to rats, and the animals in the 10mg/kg and 60mg/kg groups showed an increase in food intake and weight, and a reversible increase in the weight of the heart, liver, brain and adrenal glands. Dogs were given continuous intravenous administration for 3 months, and when the dose reached 30mg/kg, symptoms such as vomiting and salivation occurred immediately after administration. Appearance
This product is a colorless or almost colorless clear liquid.
Indication
Used for vomiting caused by radiotherapy with cytotoxic drugs.
Usage and Dosage
10 mg once a day (1 vial at a time), diluted with 40 ml of normal saline, slowly injected intravenously 30 minutes before chemotherapy.
Adverse Reactions
Some patients experience thirst, constipation, headache, dizziness, abdominal discomfort, etc. The above reactions are mild and do not require special treatment. In severe cases, there have been reports of anaphylactic shock. Therefore, the patient's reaction should be closely observed during use. If abnormalities occur, the drug should be discontinued immediately and appropriate treatment should be given.
Precautions
1. Patients who are allergic to this type of drug (5-HT3 receptor blocker) and this product are contraindicated. 2. Patients with gastrointestinal obstruction are contraindicated.
Special Population Medication
Precautions for children: The safety of the drug for children has not been determined. Precautions for pregnancy and lactation: Pregnant women should not use unless necessary. Lactating women should stop breastfeeding when using this product. Precautions for the elderly: This product is mainly excreted from the kidneys. Since the elderly often have reduced renal function, the blood concentration will continue to be high, and side effects such as headaches may occur. Therefore, the drug should be administered according to the patient's condition. If side effects occur, the dose should be reduced (for example, 5 mg).
Drug Interactions
1. This product will become turbid or crystallize when used with alkaline injection (such as furosemide, methotrexate, fluorouracil, piretanide, etc.) or etoposide injection, so it is necessary to dilute this product with sodium chloride injection before using it with the above alkaline drugs. 2. This product may reduce the content of this product when used with etoposide injection or fluoxetine sodium injection, so it should be used within 6 hours after preparation. 3. This product will become turbid or produce precipitation when used with diazepam injection, so it should be avoided to use it with them.
