pharmacological action

Animal experiments have proved that it has the functions of anti inflammation, anti exudation, increasing vein tension, improving blood circulation and correcting brain dysfunction. It has obvious inhibitory effect on experimental toe edema of rats caused by protein, carrageenan and glucan, and granuloma caused by cotton ball or formaldehyde filter paper. It has obvious protective effect on animals with brain edema caused by carbon monoxide. The experiment showed that administration 16 hours before the occurrence of edema had the highest inhibition rate of inflammation or swelling. Its inhibition effect on egg white arthritis in rats was 4-5 times that of hydrocortisone. It had significant inhibition effect on the acute exudation of croton oil in rats and the increase of capillary permeability in mice caused by histamine phosphate. Its intensity was about 7 times and 8 times that of hydrocortisone, respectively, It can also resist the exudation of ultraviolet light erythema and the increase of rabbit hind limb lymphatic permeability caused by bradykinin, and increase the tension of human vein in vitro. Intravenous administration of the drug can constrict the mesenteric veins of rats and frogs, significantly improve local blood circulation, and correct the abnormal EEG and the decrease of total brain impedance caused by brain edema in rabbits.

Pharmacokinetics

Tritium labeled sodium aescinate was administered intravenously to rats at the dose of 1mg/kg body weight, showing that it was distributed in blood, liver, kidney, muscle, stomach, intestine, salivary gland, etc. One hour after intravenous injection, about 1/3 of the dose was excreted from bile or urine, most of which was excreted by bile. After 6 hours, the blood content was only 6% of the dose.

Indications

It is applicable to traumatic swelling, brain contusion, brain concussion, cerebrovascular accident or brain edema caused by brain dysfunction in encephalitis, as well as spinal pain, varicose veins, thrombophlebitis, lower limb ulcers, swelling of anal soft tissue, burns and scald edema.

Contraindication

It is forbidden for the first three months of pregnancy. 2. Traumatic patients with renal injury, renal failure and renal insufficiency are prohibited.

matters needing attention

It should be used with caution for patients with renal failure complicated by extensive soft tissue injury, severe head injury or extensive burns.

Adverse reactions

Some patients had allergic rash, anorexia, swelling and pain at the injection site and induration after administration. It is necessary to monitor the changes of renal function when using this product. If the renal function is slightly abnormal, the drug should be stopped immediately.

Usage and dosage

Intravenous injection: 5-10mg per day. Dissolve sodium aescinate in 5~10ml of 0.9% sodium chloride injection and inject it intravenously. 1. Crisis patients, such as brain trauma and cerebral hemorrhage, can be given multiple times, but the daily dose should not exceed 25mg. 2. For patients with phlebitis tendency and women with oral contraceptives, the drug should be dissolved with 10-20ml solvent and then injected intravenously.

Drug interaction

When using sodium aescinate, other drugs that can combine with plasma protein should be used less or with caution. It is also inappropriate to use drugs with greater renal toxicity.
