Mechanism of Action

Afoxolaner acts as an antagonist at gamma-aminobutyric acid (GABA) receptors and other ligand-gated chloride channels. By blocking the transfer of chloride ions across cell membranes, it causes hyperexcitation of the insect's central nervous system, leading to death. This mechanism is selective for insects and acarines due to differences in GABA receptor sensitivity between these organisms and mammals, minimizing toxicity risks to pets.

Pharmacokinetics

After oral administration in dogs, afoxolaner exhibits high systemic absorption with a bioavailability of approximately 74%. The peak plasma concentration occurs within 2 to 4 hours post-administration. The drug has a terminal half-life of about two weeks, although this can vary significantly among different breeds.

Indications

Afoxolaner is effective against adult fleas and several tick species, including:

Fleas: Ctenocephalides felis

Ticks: Ixodes scapularis (black-legged tick), Dermacentor variabilis (American dog tick), Amblyomma americanum (Lone Star tick), and Rhipicephalus sanguineus (brown dog tick)

It also prevents flea infestations by killing fleas before they can lay eggs.

Administration

The recommended dosage for afoxolaner is typically 2.5 mg/kg body weight, administered once monthly. It can be given with or without food, and it is crucial that the entire dose is consumed by the dog. If vomiting occurs within two hours of administration, a full dose should be given again.
