    Description

    Ondansetron is a highly selective serotonin 5-HT3 receptor antagonist, with low affinity for dopamine receptors. It is effective in the treatment of nausea and vomiting caused by cytotoxic chemotherapy drugs, including cisplatin, and has reported anxiolytic and neuroleptic properties. in vitro studies confirm that Ondansetron inhibits AChE and BChE by non-competitive and mixed inhibition, respectively, with IC50 values 33 µM (AChE) and 2.5 µM (BChE). Ondansetron showed activity for preventing of ovarian hyperstimulation syndrome.

    1. General Information

    Ondansetron hydrochloride is a widely used anti - emetic (a drug that helps prevent vomiting) drug. Chemically, it is a serotonin 5 - HT₃ receptor antagonist.

    2. Mechanism of Action

    It works by blocking the action of serotonin, a neurotransmitter, at the 5 - HT₃ receptors located in the chemoreceptor trigger zone (CTZ) and on the vagal nerve terminals in the gastrointestinal tract. When chemotherapy drugs, radiation therapy, or other stimuli trigger the release of serotonin from the enterochromaffin cells of the small intestine, it binds to 5 - HT₃ receptors. Ondansetron hydrochloride inhibits this binding, thereby reducing the stimulation of the vomiting center in the brain and alleviating nausea and vomiting.

    3. Pharmacokinetics

    Absorption: Ondansetron hydrochloride is well - absorbed from the gastrointestinal tract after oral administration. The bioavailability of the oral formulation is about 56 - 75%.

    Distribution: It is widely distributed throughout the body and crosses the blood - brain barrier to reach the central nervous system (CNS), which is important for its anti - emetic effect on the CNS - mediated vomiting reflex.
