Description	Tildipirosin (TIP) is a novel 16-membered-ring macrolide authorized for the treatment of bovine and swine respiratory disease. Tildipirosin is developed to treat bacterial pathogens, including Mannheimia haemolytica and Pasteurella multocida, that cause respiratory tract infections in cattle and swine. Tildipirosin inhibits protein synthesis in bacteria and blocks the production of biofilms. Tildipirosin is particularly effective against Gram-negative pathogens.

Tildipirosin is a semisynthetic 16-membered-ring macrolide antibiotic used primarily in veterinary medicine for the treatment and prevention (metaphylaxis) of respiratory diseases in cattle and swine.

Mechanism of Action

Tildipirosin inhibits bacterial protein synthesis by selectively binding to the 23S rRNA of the 50S ribosomal subunit, blocking the elongation of the peptide chain. This action is generally bacteriostatic but can be bactericidal against certain pathogens. The drug's tri-basic character due to amine substituents on its macrolide ring contributes to its pharmacological properties.

Applications

Swine Respiratory Disease (SRD): Effective against Actinobacillus pleuropneumoniae, Pasteurella multocida, Bordetella bronchiseptica, and Haemophilus parasuis.
Bovine Respiratory Disease (BRD): Targets Mannheimia haemolytica, Pasteurella multocida, and Histophilus somni.
Used both for treatment and metaphylaxis (control in high-risk groups) of respiratory infections.
Administered as a single-dose injection: subcutaneously in cattle and intramuscularly in swine.

Safety and Side Effects

Very common side effects in pigs include immediate pain upon injection, injection site swelling, and reactions that may last several days but resolve completely.
Rare but serious adverse events like anaphylaxis and lethargy have been reported.
Toxicology studies indicate cardiovascular effects following accidental self-injection in humans; immediate medical attention is advised in such cases.
Should not be mixed with other veterinary medicinal products due to lack of compatibility data.
Not recommended for use in animals with hypersensitivity to macrolides or excipients like citric acid monohydrate or propylene glycol.

Routes of Administration

Cattle: Subcutaneous injection, usually in the neck region.
Swine: Intramuscular injection.
Single-dose administration at 4 mg/kg body weight.

Pharmacokinetics

Rapid absorption after administration with peak plasma concentrations reached within minutes in mice and somewhat longer in cattle.
Extensive distribution to lung tissue where it accumulates and persists, contributing to its long duration of action.
Long elimination half-life supports single-dose efficacy.
The PK/PD index best correlating with efficacy is the ratio of the area under the unbound concentration-time curve over 24 hours to the minimum inhibitory concentration (fAUC0-24/MIC).
Recommended clinical dose is typically 4 mg/kg body weight.
