Carbonic anhydrase is an enzyme that facilitates the reversible hydration of carbon dioxide (CO2) to form bicarbonate (HCO3-) and protons (H+). This enzymatic reaction is essential for the regulation of pH balance, the transport of carbon dioxid*e in the blood, and the maintenance of electrolyte balance in various tissues and organs.

There are several isoforms of carbonic anhydrase, each with specific localization and functions within the body. For example, carbonic anhydrase II is found in red blood cells and plays a critical role in catalyzing the rapid conversion of carbon dioxide to bicarbonate and protons, facilitating the transport of carbon dioxide from peripheral tissues to the lungs for elimination.

In addition to its role in gas exchange and pH regulation, carbonic anhydrase is involved in various physiological processes, including electrolyte and fluid secretion in the kidneys, the secretion of gastric acid in the stomach, and the production of cerebrospinal fluid in the brain.

Furthermore, carbonic anhydrase has been targeted in the development of pharmaceutical agents for the management of conditions such as glaucoma, where inhibitors of carbonic anhydrase are used to reduce intraocular pressure.

Application

Carbonic anhydrase from bovine erythrocytes (BCA) has been used to study the effect of removal of enzyme-bound metal ion, Zn2+, on aggregation behavior of the enzyme. Removal of metal ion by a chelator such as EDTA enhances the propensity of the enzyme to adopt the molten-globule state. This state of the enzyme is found to bind to the chaperone-like α-crystallin and prevent aggregation. The enzyme from Sigma has been immobilized to electrochemical transducers in order to obtain develop an analytical device for dissolved CO2 measurement. It has also been used for the thermodynamic analysis of conformational changes in BCA.
