 Located on chromosome X, HGNC is numbered 7139. Forkhead Box O4 is a Class O member of the winged spiral/forkhead transcription factor family. It was found to be a novel prostate cancer metastasis suppressor gene that may act by inhibiting PI3K/AKT and AR signaling pathways.

  FUNCTION:

• Involved in growth and differentiation: The proteins encoded by FOXO4 are regulated by factors involved in growth and differentiation, suggesting that they play a role in these processes.

• Associated with leukemia: homologues involving the translocation of this gene on the X chromosome and the Drosophila trithorax gene (encoding a DNA-binding protein) located on chromosome 11 are associated with leukemia.

Tumor suppression: When activated, Forkhead family proteins, including FOXO4, inhibit tumors by promoting cell cycle arrest and apoptosis.

Prostate cancer metastasis inhibition: FOXO4 is a novel prostate cancer metastasis suppressor gene that may act by inhibiting PI3K/AKT and AR signaling pathways.

• Regulation of other genes: FOXO4 can regulate the expression of other genes, such as RUNX2, so as to exert its biological function.

• Binding and inhibiting proteins: FOXO4 binds and inhibits myocardin, thereby inhibiting smooth muscle cell differentiation. The protein levels of HIF-1α, a key protein in hypoxic reactions, can also be down-regulated.
