Cagrilintide (AM833, NN0174-0833) is a long-acting, lipidated amylin analogue developed by Novo Nordisk for the research application of obesity and type 2 diabetes. It is a 39-amino-acid peptide based on the in vivo amylin sequence, with modifications to reduce β-sheet propensity and amyloid fibril formation, a C-terminal proline amidation, and a disulfide bridge between Cys3 and Cys8. A C20 fatty diacid side chain is attached via a γ-Glu linker at the N-terminus, enabling albumin binding and extending the plasma half-life to approximately 1 week, supporting once-weekly research route application.
