Fezolinetant (CAS 1629229-37-3), also known as ESN364, is an orally bioavailable, non-peptide neurokinin-3 receptor (NK3R/TACR3) antagonist with the molecular formula C₁₆H₁₅FN₆OS and a molecular weight of 358.39 g/mol . The compound features a unique fused heterocyclic core containing a triazolo[4,3-a]pyrazine ring system, substituted with a 4-fluorophenyl methanone group, a methyl group at the 8-position, and a 3-methyl-1,2,4-thiadiazol-5-yl moiety . Fezolinetant is approved for the treatment of moderate-to-severe vasomotor symptoms (hot flashes) associated with menopause.
