Rifabutin Use and Manufacturing

Methods of Manufacturing

Using 1, 4-dihydrorifamycin S as raw material, rifabutin can be obtained by bromination, nitroso substitution, hydrogenation, amination, and then reacting with N-isobutylpiperidone.

Uses

Rifamycins are antibiotics that inhibit DNA-dependent RNA polymerases and are usually bactericidal against Gram-positive bacteria but bacteriostatic against Gram-negative bacteria. Rifamycins are also effective against Mycobacterium species, including M. tuberculosis. Rifabutin is a broad-spectrum rifamycin antibiotic that has applications against tuberculosis, H. pylori, M. avium complex, Chlamydia, and other bacteria. It is also useful in co-infections with human immunodeficiency virus, including tuberculosis.

Drug Function and Efficacy

This product is a semi-synthetic rifamycin drug with similar structure and activity to rifampicin. In addition to its anti-Gram-negative and anti-Gram-positive bacteria effects, it also has anti-tuberculosis and avian Bacillus (M.avium) activity. Recent studies have shown that in HIV-infected lymphocytes, the use of 0.1μg/ml of this product has an inhibitory effect on 92% of reverse transcriptase.
