Cephalins, brain structure

Cephalins, brain

structure

Description

Derivatives of phosphatidic acids in which the phosphoric acid is bound in ester linkage to an ethanolamine moiety. Complete hydrolysis yields 1 mole of glycerol, phosphoric acid and ethanolamine and 2 moles of fatty acids.

Cephalins, brain Use and Manufacturing

Methods of Manufacturing

Method 1. Alcohol-ether separation and extraction of cholesterol and lecithin. Take fresh or frozen brain or spinal cord, remove membranes and bloodshot impurities, mince, immerse in 1.2 times the amount of acetone for 4.5h, continuously stir, filter, and immerse 5 times repeatedly, filtrate Make cholesterol. The filter cake is vacuum dried, 2-3 times the amount of ethanol is added, and the mixture is extracted at 35-40°C for 12 hours, filtered, and then extracted once. The filtrate is made into lecithin, and the filter cake is vacuum dried. Preparation of the precipitate: Add ether to the filter cake and extract for 12 hours, filter, and extract once more. Combine the two filtrates and concentrate the filtrate to obtain a concentrate. Place the concentrated solution at 0°C for 12 hours, centrifuge, take the supernatant and add ethanol at 60°C, stir while adding, filter, and collect the precipitate. Filter cake [ether]→[12h] filtrate [concentration, centrifugation]→supernatant [ethanol]→precipitate preparation of cephalin finished product Add diethyl ether to the precipitate, stand at 0℃, and filter to obtain the filtrate. Add ethanol to the filtrate, precipitate, filter, collect the precipitate, and vacuum dry to obtain the finished cephalin. Precipitate [ether] → [0℃] filtrate [ethanol] → finished cephalin. Method 2. Preparation of crude cephalin by calcium chloride precipitation. Take the dry brain and add 3 times the amount of ether, heat to boiling, filter, and then extract the residue Once, combine the two filtrates and concentrate the filtrate to 1/3 volume to obtain a concentrated solution. Dried brain [ether] → filtrate [concentration] → preparation of concentrated liquid crude cephalin Add 3 times more than 80% ethanol to the concentrated liquid, heat to dissolve in a water bath, cool to overnight, layer, filter, filtrate to make eggs Phospholipids and cholesterol, precipitate with a small amount of water, add 5g/L. Calcium chloride, collect the precipitate at 100°C, and dry the precipitate to obtain crude cephalin. Preparation of finished cephalin product The crude cephalin is added with 3 times the amount of ethanol, the cold room is precipitated for 4 hours, filtered, and this is repeated 5 times to obtain the precipitate, and the precipitate is dissolved with ether, filtered to obtain the filtrate, and the filtrate is added with the same amount of ethanol or acetone to precipitate, Filter and vacuum dry the precipitate to obtain the finished cephalin. Crude cephalin [ethanol]→precipitation [ether]→filtrate [ethanol or acetone]→cephalin finished crude cephalin [ethanol]→precipitation [ether]→filtrate [ethanol or acetone]→cephalin finished product.
