Bacitracin Use and Manufacturing

Methods of Manufacturing

The producing bacterium is Bacillus subtilis. The medium is defatted soybean flour (5%), sucrose (1.2%), ammonium sulfate (0.2%), calcium carbonate (0.2%), etc. The cultivation process of bacteria can be divided into two stages: bacterial development and antibiotic production. The optimal temperature for producing bacitracin is about 37℃, while the temperature during the bacterial development period needs to be slightly higher than 1-2℃, and the ventilation per minute is 90% of the fermentation broth. The bacitracin produced is adsorbed on the carboxylic acid type exchange resin under a pH value of 4-5.7, and then eluted with a weak base (ammonia).

Uses

Bacitracin is a peptide antibiotic effective against gram-postive bacteria. Bacitracin is an inhibitor of peptidoglycan synthesis. Bacitracin disrupts bacterial cell wall synthesis by inhibiting dephosphorylation of lipid pyrophosphate. Bacitracin also strongly inhibits proline endopeptidase from human muscle.

Drug Function and Efficacy

It has a strong antibacterial effect on Gram-positive bacteria such as Staphylococcus and Streptococcus. It also has a certain effect on Neisseria gonorrhoeae, Neisseria meningitidis and some spirochetes and actinomycetes. All Gram-negative bacteria are resistant. Systemic medication can cause severe nephrotoxicity, so it is currently limited to local application.
