Methylthiouracil is an antithyroid agent. Methylthiouracil suppresses the production TNF-α and IL-6, and the activation of NF-κB and ERK1/2.

As stipulated in 40 CFR 261.33, when methylthiouracil, as a commercial chemical product or manufacturing chemical intermediate or an off-specification commercial chemical product or a manufacturing chemical intermediate, becomes a waste, it must be managed according to Federal and/or State hazardous waste regulations. There are no known natural sources of methylthiouracil(1).
Release of methylthiouracil to the environment may occur as a result of its production and use as a drug that has been used in the past to treat human hyperthyroidism(1,2). As of 1970, the compound was more broadly used outside the U.S(2). No evidence was found that indicates that methylthiouracil has ever been produced commercially in the U.S.(2,SRC).
TERRESTRIAL FATE: If methylthiouracil is released to soil, it will be expected to be highly mobile in soil(1,SRC) based upon an estimated Koc of 138 which was calculated(2,SRC) from a water solubility of 533.2 mg/l at 25 °C(3) using a recommended regression equation(2,SRC). It may, therefore, leach to groundwater(1,SRC). It is not known whether the compound will photooxidize or biodegrade in soil or volatilize from moist near-surface soil(SRC). But it may slowly biodegrade in natural waters and soil based upon the slow biodegradation of the structurally similar 2-thiouracil by activated sludge from municipal treatment plants(4). Methylthiouracil should not be susceptible to hydrolysis in water based upon a measured neutral hydrolysis rate constant of 9.7X10-6 hr-1 at 25 °C which corresponds to a half-life of 8.8 years(5).|AQUATIC FATE: If it is released to water, methylthiouracil may be susceptible to direct photolysis in surface water based upon its absorption of light at wavelengths greater than 290 nm in methanol solution(1). The presence or absence of any of these forms may affect the reactivity of the compound in the environment(SRC). Methylthiouracil may be susceptible to direct photolysis in water and in the atmosphere based upon the absorption of light at wavelengths greater than 290 nm by methylthiouracil in methanol solution(3).|Methylthiouracil will be expected to be susceptible to photooxidation in the atmosphere based upon estimated rate constants for the vapor phase reactions of two of its possible tautomeric forms with photochemically produced hydroxyl radicals and ozone(1,2,SRC). The rate constant for the vapor phase reaction of the oxo-thiourea form of the compound with photochemically produced hydroxyl radicals has been estimated to be 100X10-12 cu cm/molecule-sec at 25 °C which corresponds to an atmospheric half-life of 3.8 hr at an atmospheric concentration of 5X10+5 hydroxyl radicals per cu cm(1,SRC). The rate constants for the vapor phase reactions of the hydroxy-sulfhydryl-pyrimidine form of the compound with photochemically produced hydroxyl radicals and ozone have been estimated to be 168X10-12 cu cm/molecule-sec and 1.14X10-17 cu cm/molecule-sec at 25 °C, respectively, which correspond to an overall atmospheric half-life of 2.1 hr at atmospheric concentrations of 5X10+5 hydroxyl radicals per cu cm and 7X10+11 ozone molecules per cu cm(1,2,SRC).|Methylthiouracil should not be susceptible to hydrolysis in water or moist soil based upon a measured neutral hydrolysis rate constant of 9.7X10-6 hr-1 at 25 °C which corresponds to a half-life of 8.8 years(1).

By the reaction of ethyl acetoacetate and thiourea. Dissolve thiourea in sodium hydroxide solution, add ethyl acetoacetate dropwise, and control the temperature below 37°C. After 2h of reaction, water and activated carbon were added to decolorize at 70-80°C for 0.5h. Filter, the filtrate is acidified with hydrochloric acid to pH 4-5, cooled to room temperature, filtered and washed. The filter cake is recrystallized with water to obtain a finished product. The yield was 66%. In production, ethyl acetoacetate and thiourea can also be mixed first, and then sodium oxide solution can be added dropwise; anhydrous sodium carbonate can also be used instead of sodium hydroxide, and the yield is about the same.
