Lentinan is purified β-glucan from Shiitake mushrooms. Lentinan has been approved as a biological response modifier for gastric cancer in Japan[1].


Polysaccharide isolated from the edible mushroom LENTINULA EDODES. The exact composition is unknown.

Drug Information

Substances that inhibit or prevent the proliferation of NEOPLASMS. (See all compounds classified as Antineoplastic Agents.)|Substances that augment, stimulate, activate, potentiate, or modulate the immune response at either the cellular or humoral level. The classical agents (Freund's adjuvant, BCG, Corynebacterium parvum, et al.) contain bacterial antigens. Some are endogenous (e.g., histamine, interferon, transfer factor, tuftsin, interleukin-1). Their mode of action is either non-specific, resulting in increased immune responsiveness to a wide variety of antigens, or antigen-specific, i.e., affecting a restricted type of immune response to a narrow group of antigens. The therapeutic efficacy of many biological response modifiers is related to their antigen-specific immunoadjuvanticity. (See all compounds classified as Adjuvants, Immunologic.)

Methods of Manufacturing

Extracted from mushrooms. 200kg of fresh mushrooms are washed with water, crushed and mixed, and extracted with 1000L of boiling water for 8-15 hours. The resulting suspension is filtered or centrifuged to remove insoluble materials. The aqueous solution was concentrated under reduced pressure until slight turbidity appeared, and an equal volume of ethanol was added. The precipitate is collected by filtration or centrifugal separation to obtain 320 g of lentinan-containing components. Take 50g of this component and suspend it in 2L of water and mix at room temperature to form a uniform viscous brown solution. Then add 20L of water and stir for 1-2h to obtain a clear and homogeneous solution. Under vigorous stirring, 0.2mol/L CTA-OH aqueous solution (pH value 13.2) was added dropwise, and flocculent precipitation appeared at pH value 7-8, and a large amount of white precipitate appeared at pH value 10.5-11.5. Add 0.2mol/L CTA-OH dropwise until no more precipitation appears, at this time the pH value of the solution is about 12.8. Collect all the precipitates by centrifugation and wash them with ethanol. It was suspended in 1.2L of 20% acetic acid and stirred at 0°C for 5 min. The precipitate was collected by filtration, and stirred with 1L of 50% acetic acid at 0°C for 3min. The precipitate was collected by centrifugal separation and then dissolved in 2L of 6% sodium hydroxide aqueous solution. After centrifugal separation to remove impurities, 4L ethanol was added. The precipitate that appeared was collected, washed twice with ethanol, then washed once with ether, and dried in vacuum to obtain a powder. It was developed by the Sevag method with chloroform and 1-butanol, and 3 times the volume of ethanol was added to precipitate it. The precipitate was collected, washed twice with methanol and once with ether, and dried under vacuum at room temperature in the presence of calcium chloride to obtain lentinan. 31g of lentinan can be extracted from 200kg of raw materials.

Uses

Used in combination with chemotherapy drugs in patients with gastrointestinal tumors, it can prolong survival time and prevent leukopenia or serum total protein reduction. It has a certain effect on chronic hepatitis B, can significantly improve the patient~"s symptoms, make the serum alanine aminotransferase normal or better, and adjust the patient~"s immune function. Note: Even anaphylactic shock, white blood cell and hemoglobin reduction symptoms may occur. Children and women of pregnancy and childbearing age should be used with caution.
