Global TOP10 pharmaceutical companies by market capitalization - Review of AbbVie R&D pipeline
AbbVie (AbbVie) announced that upadacitinib, a once-daily oral, highly selective JAK inhibitor, was approved in Japan for induction remission and maintenance in adults with moderate-to-severe ulcerative colitis who have failed to respond to conventional therapy. Abbvie is expanding once again in the field of immunity.
Since AbbVie listed on the New York Stock Exchange in 2013, its market capitalization has risen from $18 billion to more than $250 billion in the past decade. What's driving AbbVie's growth?
Split at its height
In January 2013, AbbVie officially went public on the New York Stock Exchange. But AbbVie's roots go back to Abbott laboratories, which was founded in 1888.
Headquartered in Chicago, Abbott became the world's top pharmaceutical giant back in 2010. As the bioppharmaceutical industry entered a booming period, White, then the CEO of Abbott, realized that investors had different valuations and investment intentions for Abbott's traditional business and innovative drugs, so he decided to split the company into two companies: The diversified legacy businesses will remain under Abbott, while the bioparmaceutical innovation business will be under a new AbbVie. After a year of transition and preparation, AbbVie was born on January 2, 2013, with its R&D-centered biopharmaceuticals assets officially listed on the New York Stock Exchange.
The same CEO who bought Knoll, BASF's pharmaceutical division, for $6.9 billion in 2000, acquired an investigational drug called D2E7, known today as adalimumab, which has been AbbVie's most important source of revenue since its inception.

Abbvie invested $7.08 billion in R&D in 2021, as a percentage of revenue: 12.60%. In the first half of 2022, AbbVie's total revenue was $28.121 billion, and Humilar sales were $10.099 billion, continuing to increase by 1.7%.
From the perspective of business segments, AbbVie has layouts in immunology, oncology, aesthetics, neuroscience, ophthalmology and other fields.
The immunization segment is far and away the most important segment, with revenue of $13.348 billion in the first half of 2022, up 12.5% year over year and accounting for 47.5% of total revenue. The second is the oncology segment, with business revenue of $3.296 billion in the first half of 2022; Neuroscience revenue was $3.146 billion, up 16.2% year over year; Aesthetics revenue was $2.745 billion, up 6.6% year on year; Revenue from ophthalmic products was $1.488 billion, down 14.3% year on year.

The drug component of Humele is Adalimumab, which is the world's first fully human monoclonal antibody against tumor necrosis factor (TNF-α active monoclonal antibody) approved for sale. However, the drug king also has roadblocks, and the patent cliff is the dilemma AbbVie has to face.
In Europe, the patent of Humira expired in 2018, and its sales in Europe began to decline due to the impact of similar drugs.

The United States is Humilor's main market, and its patent will expire in 2023, when the myth of "drug king" may not continue.
To compensate for the coming decline in Humira, AbbVie has launched two supernovas in the immunology space: the JAK inhibitor Rinvoq and the IL-23 inhibitor Skyrizi. Abbvie has high hopes for the two products, which are expected to generate $15 billion in sales by 2025.
upadacitinib (Rinvoq) is a selective JAK1 inhibitor that belongs to the second generation of selective JAK inhibitors. Up to now, Rinvoq has been approved globally for five indications: atopic dermatitis (AD), rheumatoid arthritis (RA), psoriatic arthritis (PsA), ulcerative colitis (UC) and active ankylosing spondylitis (AS). In China, two indications of Rinvoq were successively approved by the National Food and Drug Administration in the first half of this year for the treatment of moderate to severe atopic dermatitis and psoriatic arthritis (PsA). In the first half of 2022, Rinvoq revenue reached $1.057 billion, up 55.1% year over year.
Skyrizi (risankizumab, Risankizumab) isan IL-23 inhibitor that has been approved in the United States for moderate-to-severe plaque psoriasis, active psoriatic arthritis (PsA), and moderate-to-severe Crohn's disease (CD). Skyrizi's revenue in the first half of 2022 was $2.192 billion, up 75.7% year on year.
The fields of oncology, aesthetics, neurology and ophthalmology are blossoming
Hummel was once AbbVie's pride, but it was also a danger. With a series of acquisitions, mergers and acquisitions, AbbVie has emerged in many areas.
Oncology is AbbVie's second largest segment in addition to immunology and includes the BTK inhibitor Imbruvica (ibrutinib) and the Bcl-2 inhibitor Venclexta (Venetoclax). In the first half of 2022, revenue of the two products was $2.318 billion (-12.5%) and $978 million (+16.5%), respectively. Abbvie is also expanding its oncology pipeline, following a $3.9 billion partnership with Genmab to introduce bispecific antibody drugs epcoritamab and ABBV-383, and the acquisition of BCMA/CD3 bispecific antibody TNB-383B from TeneoOne. All are currently in clinical trials.
In 2020, AbbVie spent $63 billion to acquire Allergan, giving it access to aesthetic stars Botox and Juvederm, as well as neuroscience products Botox Therapeutics, Vraylar and Ubrelvy.
In the first half of 2022, AbbVie's neuroscience product revenue was $3.146 billion, and its aesthetics segment revenue was $2.745 billion. Among them, Botox Cosmetic and Botox Therapeutic contributed $1.336 billion and $1.292 billion, respectively. Two new migraine drugs, Ubrelvy (ubrogepant) and Qulipta (atogepant), are also coming to prominence.
Another area AbbVie is betting heavily on is ophthalmology. The current best seller is Restasis, a dry eye drug approved in 2002, with revenue of $414 million in the first half of this year, but it is facing a generic attack as its patent expires. Vuity is another important product in AbbVie's ophthalmic pipeline. In October 2021, Vuity received FDA approval for the treatment of presbyopia, making it the first eye drop specifically designed to treat presbyopia. In addition, in September 2021, AbbVie teamed up with Regenxbio to develop and commercify RGX-314, a potential one-time gene therapy for the treatment of wet age-related macular degeneration (wAMD), diabetic retinopathy (DR), and other chronic retinal diseases.
The small knot
From 2013 to now, in less than ten years, AbbVie has grown from a "small and beautiful" innovative pharmaceutical company when it was initially split to a "giant" with a market value of more than 250 billion US dollars. The disease field has also expanded from immunology to immunology, oncology, neuroscience, ophthalmology, aesthetics and other fields.
It is worth to mention that AbbVie China R&D Center, namely Shanghai Zhangjiang R&D Center, was completed as early as 2009, and AbbVie China was officially established in June 2013. Abbvie currently employs about 1,200 people in China, including more than 250 research and development personnel (nearly a quarter of all employees). Abbvie is expected to launch more than three innovative drugs and seven new indications in the Chinese market by the end of 2025, covering immunology, hematological oncology, solid tumors and other fields, and strive to promote China to become the largest overseas market outside the headquarters.
Abbvie is still expanding its pipeline in a variety of ways, including small molecules, antibodies and cell therapies. According to the previous prediction of the well-known American media Evaluate pharma, AbbVie will become the world's largest pharmaceutical company in prescription drug sales by 2028, with expected sales of $65.7 billion.
