The ADC Journey:
It is 2024, and ADCs have become highly sought-after therapies. However, Novartis, with characteristic pragmatism, explains that it is not a lack of love but a matter of compatibility that has steered its course away from ADCs. Novartis has previously invested significant effort in ADC research and development, but unfortunately, success has eluded the company. Past experiences have left a lingering sense of caution, perhaps akin to the adage "once bitten, twice shy." Nevertheless, viewed from a different perspective, Novartis' departure from the ADC arena can be seen as an astute move for a pharmaceutical giant.
The Patent Dispute and Novartis' New Frontier:
On January 17th, a long-standing patent dispute between two ADC giants, Daiichi Sankyo and Seagen, reached a temporary resolution. The U.S. Patent and Trademark Office's final decision ruled that Enhertu's patent was not infringed upon. While the dust settles on this legal battle, Novartis has chosen to forge a new path—one that encompasses a more integrated approach to therapeutics, particularly in the field of radiopharmaceuticals. At present, Novartis appears to have a stronger competitive advantage in this arena. However, this transition is not without its challenges.
Novartis' ADC Trials and Safety Concerns:
Novartis has previously explored partnerships and independent development in the ADC space. However, these endeavors faced hurdles due to safety concerns and limited efficacy. For instance, the Phase I clinical trial of CDH6 ADC drug HKT288, developed in collaboration with ImmunoGen, encountered neurological adverse reactions and lacked definitive efficacy, leading to its discontinuation. Despite these setbacks, Novartis remains cautious, as the unique structure of ADCs often accompanies substantial toxicities. In light of this, Novartis believes that radiopharmaceuticals might offer a safer alternative, particularly in comparison to certain ADCs.
The ADC Market and Novartis' Strategic Focus:
The ADC market features prominent players such as Takeda Pharmaceutical, Roche, Pfizer, AstraZeneca, Gilead, Astellas Pharma, Daiichi Sankyo, GlaxoSmithKline, and ADC Therapeutics. Experts predict that the ADC market will experience rapid growth, with a compound annual growth rate of 31.2% from 2021 to 2030, reaching a market size of $63.8 billion by 2030. Within this landscape, Kadcyla and Enhertu have emerged as leading contenders. Kadcyla, the first HER2 ADC product to be approved, has witnessed steady sales growth, reaching $2.289 billion in 2022. Enhertu, a collaboration between AstraZeneca and Daiichi Sankyo, has garnered significant attention and acclaim, with sales soaring to $1.317 billion in 2022. Analysts predict that Enhertu may surpass Kadcyla and become the flagship ADC by 2025.
Novartis' Strategic Pivot:
In the midst of this transformative era, Novartis has chosen to exit the ADC arena and redirect its focus to its strengths, particularly in radiopharmaceuticals. Radiolabeled targeted therapies, known as Radiopharmaceuticals Development Candidates (RDCs), have become Novartis' priority. By leveraging the power of precision targeting and radiation, Novartis aims to revolutionize the field of nuclear medicine. This strategic pivot allows Novartis to navigate uncharted territory and tap into the potential of RDCs, positioning itself as an innovative leader in the pharmaceutical industry.
