Debut Biotechnology Creates Animal-Free Replica of Carmine
Carmine (also known as Natural Red 4, CI 75470 or E120) is a deep red pigment widely used in cosmetics, food and textiles. It is made from carminic acid, which is extracted from a variety of insect species, including the cochineal insect. That may soon change!
Debut Biotechnology, a startup based in San Diego, California, has announced a real breakthrough in the development of animal-free carmine. Thanks to a novel enzyme, they were able to bio-manufacture a vegan version of the deep red pigment, which until now has been sourced from the cochineal insect.
“Carmine is one of the most difficult molecules to innovate. After significant investment and hard work, we are very proud to have discovered the most important missing step in creating renewable carmine, allowing us to go from sugar all the way to this precious molecule,” said Dr. Joshua Britton, Founder and CEO of Debut.
To enable bioproduction from renewable, low-cost sources, Debut combined its patented microbial production system with cutting-edge cell-free biomanufacturing to overcome industrial limitations and increase bioproduction rates 100-fold, which was unattainable with previous best-in-class attempts. This required Debut to discover two enzymes and improve their performance to break through known barriers in the bioproduction of carmine.
According to Debut, existing synthetic alternatives to carmine are imprecisely color matched and lack the vividness, stability, and formulation potential of animal-derived carmine. There are also concerns about synthetic alternatives. In January 2025, the FDA banned the use of synthetic Red No. 3 due to claims that the additive is carcinogenic.
In contrast, Debut claims that its biomanufactured carmine has "unparalleled health, safety, animal-free, and environmental advantages, in addition to ensuring a consistent supply chain that is not dependent on beetle migration patterns." In addition, Debut's carminic acid is more than 95% pure, while the industry average is 30% or less. Carminic acid extracted from cochineal insects is available in very low purity, typically 10%.
Biomanufactured carmine also does not contain a specific protein found in beetle carmine that is known to cause skin irritation, making it ideal for beauty and food applications. In addition, biotechnologically derived carmine has antioxidant and anti-inflammatory properties.
Debut recently completed pilot-scale bioproduction, enabling the formulation of carmine acid for beauty applications. As a first step, this will enable the company to supply carmine beauty products, an industry first, followed soon by food and beverage applications.
Last July, the California-based startup announced that it had signed an agreement with L’Oréal to develop a dozen key bioidentical ingredients that will replace conventionally sourced ingredients currently used in skin and hair care products, makeup and fragrances sold globally by the beauty giant. In January of this year, Debut also announced a partnership with contract manufacturer KDC/one to rapidly create entirely new ingredients through computational compound predictions.
