Function and Efficacy
Pharmacological action: Minocycline is a semi-synthetic tetracycline antibiotic with antibacterial activity against both Gram-positive and Gram-negative bacteria. Its antibacterial spectrum is basically the same as that of tetracyclines. The mechanism of action of erythromycin is to exert its antimicrobial effect by inhibiting protein synthesis. This product is sensitive to sensitive Gram-positive bacteria, including Staphylococcus aureus, Streptococcus, and Neisseria meningitidis; it is relatively sensitive to Gram-negative bacteria, including gonococci and Haemophilus influenzae; it has a weaker effect on Gram-negative bacilli; this product also has a certain effect on microorganisms such as Chlamydia trachomatis and Ureaplasma. Indication
It is suitable for the following infections caused by bacteria such as Staphylococcus, Streptococcus, Pneumococcus, Gonorrhea, Shigella, Escherichia coli, Klebsiella pneumoniae, Enterobacter aerogenes, Bacillus anthracis, Klebsiella, Proteus, Actinomyces and pathogenic microorganisms such as Chlamydia, Mycoplasma, Rickettsia, Treponema pallidum, etc. that are sensitive to this product: 1. Sepsis and bacteremia. 2. Superficial suppurative infections: folliculitis, pyoderma, furunculosis, carbuncle, cellulitis, sweat gland inflammation, acne, sebaceous cyst atheroma, papillary dermatitis, paronychia, abscess, secondary infection of corns, tonsillitis, periarthritis of shoulder, pharyngitis, dacryocystitis, blepharitis, sty, gingivitis, pericoronitis, dental maxillary sinusitis, infectious maxillary cyst
Usage and Dosage
Oral administration, the dosage is as follows: 1. Adults: The usual dosage of this product is 100 mg every 12 hours. Alternatively, when the frequency of administration needs to be increased, it can be taken 4 times a day, 50 mg each time. Disseminated acne: 100 mg per day, either as a single dose or in 2 doses, the course of treatment depends on the drug response. Non-gonococcal urethritis: 100 mg per day, either as a single dose or in 2 doses, the course of treatment is 10-14 days. Gonorrhea: For male patients, the initial dose is 200 mg, followed by 100 mg every 12 hours, for at least 4 days of treatment, and post-treatment culture within 2-3 days; the dosage for female patients is the same as that for men, and treatment is required for 10-14 days. Systemic infection: The usual dose is 1 every 12 hours.
Adverse Reactions
1. Digestive system: anorexia, nausea, vomiting, diarrhea, glossitis, dysphagia, enterocolitis, pancreatitis and inflammatory lesions in the anogenital area (excessive growth of Candida), increased liver enzymes, and rare cases of hepatitis. Esophagitis and esophageal ulcers have occurred in a few patients. Most of these patients take the medicine at bedtime. 2. Skin reactions: maculopapular rash, erythematous rash, rarely erythema multiforme, Stevens-Johnson syndrome and exfoliative dermatitis, as well as photosensitivity reactions. Cases of skin and mucous membrane pigmentation have been reported. 3. Renal toxicity: Increased BUN has been reported, and it is clearly dose-related. 4. Allergic reactions: urticaria, angioedema, polyarticular pain
Precautions
It is contraindicated for patients who are allergic to tetracyclines.
Special Population Medication
Precautions for children: Because this product can cause permanent discoloration of teeth, enamel hypoplasia, and inhibit bone development and growth, it is not recommended for children under 8 years old unless the expected benefits outweigh the possible risks. Precautions for pregnancy and lactation: 1. Animal research results show that tetracyclines can pass through the placenta and can be found in fetal tissues, causing toxic effects on fetal development (usually associated with delayed bone development). Treatment in early pregnancy in animals can show signs of embryotoxicity. 2. Minocycline hydrochloride, like other tetracycline antibiotics, can pass through the placenta and cause fetal damage when taken by pregnant women. If minocycline hydrochloride is taken during pregnancy or if a woman becomes pregnant while taking the drug, the patient should be informed of the potential risks of the drug to the fetus. 3. The use of tetracyclines during tooth development (late pregnancy) can cause permanent discoloration of teeth. Incomplete development of enamel has also been reported. 4. Taking tetracyclines in the last three months of pregnancy can form stable calcium complexes in fetal bones. A decrease in fibula growth rate can be observed in immature infants and young children taking tetracycline drugs (25 mg/kg every 6 hours). Changes in fibula growth rate can be restored after discontinuation of the drug. 5. Congenital malformations, including limb reduction, have been reported in post-marketing clinical experience. 6. Minocycline hydrochloride can be secreted in human milk; therefore, a decision should be made whether to stop breastfeeding or stop taking the drug. Precautions for the elderly: Clinical trials of this product did not include enough patients over 65 years old, so it is impossible to determine whether the response of the elderly after taking the drug is the same as that of young people. The dose selection for elderly patients should be cautious, usually starting with the minimum dose, because the elderly are more likely to have reduced liver, kidney or heart function, and may also suffer from other diseases or be taking other drugs.
Drug Interactions
1Since tetracyclines can inhibit prothrombin activity, patients treated with anticoagulants should reduce the dose of anticoagulants. 2Since tetracyclines are antibacterial agents that can interfere with (hinder) the bactericidal effect of penicillin, they should be avoided in combination with penicillin. 3Calcium, magnesium, aluminum, and iron preparations may reduce drug absorption when used in combination with this drug. 4The combination of tetracyclines and methoxazole can cause severe nephrotoxicity. 5The simultaneous use of tetracyclines can reduce the efficacy of oral contraceptives.
