Dacarbazine for injection
Function and Efficacy
1 Pharmacology: This product has anti-tumor effects. Since it is a precursor of purine biosynthesis, it can interfere with purine biosynthesis. After entering the body, it demethylates in the liver microsomes to form monomethyl compounds, which have direct cytotoxic effects. It mainly acts on the G2 phase. It inhibits the synthesis of purine, RNA and protein, and also affects the synthesis of DNA. Therefore, it is also considered to be an alkylating anti-tumor drug. 2 Toxicology: The experimental results show that the acute median lethal dose (LD50) of this product for mice injected intraperitoneally is 815mg/kgplusmn;50.9mg/kg.

Indication
This product is used to treat malignant melanoma
Usage and Dosage
1. Intravenous injection. 200-400mg/m2 per day, for 5-10 consecutive days. To reduce irritation to blood vessels, use 10-15ml of normal saline, dissolve it, dilute it with 250ml of 5% glucose solution and then drip it. It takes more than 30 minutes to drip it. One course of treatment is 5-10 days of continuous use. Generally, the drug is repeated every 3-6 weeks. For malignant melanoma of the limbs, arterial injection at a nutritious dose can be used. The blood drug concentration reaches its peak 30 minutes after each intravenous injection, drops to zero within 6 hours, and 30% is excreted in the urine within 0-6 hours. It cannot pass the blood-brain barrier. Single large dose: 650-1450mg/㎡, once every 4-6 weeks. 2. Intravenous drip: 200mg/㎡ each time, once a day, for 5 consecutive days, and repeat the medication every 3-4 weeks. 3. Arterial infusion: arterial infusion
Adverse Reactions
1. Gastrointestinal reactions: such as loss of appetite, nausea, vomiting, diarrhea, etc., which may be alleviated or disappear after 2 to 8 hours. 2. Bone marrow suppression: It may cause a decrease in white blood cells and platelets, and anemia, which is more obvious at high doses. Generally, a decrease in blood count occurs 2 to 3 weeks after medication, and it may return to normal in the 4th to 5th week. 3. A small number of patients may experience "flu"-like symptoms, such as general discomfort, fever, and muscle pain, which may occur 7 days after medication and last for 1 to 3 weeks. Facial numbness and hair loss may also occur. 4. Local reactions: There may be vascular irritation reactions at the injection site. 5. Occasionally, liver and kidney damage may occur.
