Azasetron Hydrochloride Tablets

Function and Efficacy

This product is an antiemetic drug of the 5-hydroxytryptamine type 3 (5-HT3) receptor antagonist class. It has a strong and selective antagonistic effect on 5-HT3 receptors, but no dopamine receptor antagonism. This product has a high affinity for 5-HT3 receptors, and its action strength is about 410 times stronger than that of metoclopramide, 2 times that of ondansetron, and almost equal to granisetron. After intravenous injection of this product into the body, it can highly antagonize bradycardia induced by 5-hydroxytryptamine (5-HT) (i.e., Bezold=Jarisch reflex, which is a manifestation of the excitatory effect of 5-HT3 receptors). The antagonistic effect of this product is about 900 times stronger than that of metoclopramide, about 4 times stronger than that of ondansetron, and equal to granisetron. In addition, in in vitro tests on sensory nerve cells, isolated hearts or isolated ileum, this product showed an antagonistic effect on 5-HT3 receptors, which was 20 to 720 times stronger than metoclopramide and 1/8 to 3 times stronger than graceron.

Indication

It is suitable for the treatment of gastrointestinal symptoms such as nausea and vomiting caused by taking anti-malignant tumor drugs (cisplatin, etc.).

Usage and Dosage

The usual dose for adults is: intravenous injection, 10 mg once a day.

Adverse Reactions

1. Nervous system: Occasionally headache, heaviness, anxiety, irritability. 2. Digestive system: Occasionally thirsty. 3. Circulatory system: Occasionally pale face, cold feeling, palpitations. 4. Liver: Occasionally elevated GOT and GPT values. 5. Occasionally rash on the skin. 6. Others: Occasionally fever, chills, fatigue, cramps in both feet.

Precautions

This product is mainly excreted from the kidneys. Due to the decreased renal function of the elderly, the continuous high blood concentration after using this product may easily cause adverse reactions such as headache and heaviness of the head. Therefore, the elderly should pay attention to their condition when using it. If adverse reactions occur, the dosage should be reduced (for example, to 5 mg).

Drug Interactions

This product is prone to turbidity or crystal precipitation when mixed with alkaline injections such as furosemide injection, methotrexate injection, and fluorouracil injection, so it should be mixed with normal saline for use.
