Function and Efficacy

Antiviral drug. It has an inhibitory effect on herpes simplex virus, varicella zoster virus, cytomegalovirus, etc. in vitro. After entering the cells infected by herpes virus, this product competes with deoxynucleoside for viral thymidine kinase or cell kinase, and the drug is phosphorylated into activated acyclovir triphosphate, and then inhibits viral replication in two ways: ① Interfering with viral DNA polymerase to inhibit viral replication; ② Under the action of DNA polymerase, it binds to the growing DNA chain, causing the extension of the DNA chain to be interrupted. This product has a special affinity for viruses, but has low toxicity to mammalian host cells. There are reports of carcinogenicity in in vitro cell transformation assays, but no evidence of carcinogenicity has been found in animal experiments. Some animal experiments show that high concentrations of drugs can cause mutations, but there is no evidence of chromosomal changes. The carcinogenic and mutagenic effects of this product are still unclear. High-dose injection can cause testicular atrophy and reduced sperm count in animals. The drug can pass through the placenta, and animal experiments have confirmed that it has no effect on the embryo.

Appearance

Indication

1. Herpes simplex virus infection: used for the initial and recurrent cases of genital herpes virus infection. For recurrent cases, this product can be taken orally for prevention. 2. Herpes zoster: used for the treatment of herpes zoster in people with normal immune function and mild cases in people with immunodeficiency. 3. The treatment of chickenpox in people with immunodeficiency.

Usage and Dosage

Adverse Reactions

Occasionally, dizziness, headache, joint pain, nausea, vomiting, diarrhea, stomach discomfort, loss of appetite, thirst, leukocytopenia, slight increase in proteinuria and urea nitrogen, skin itching, etc. may occur. Acne, insomnia, and menstrual disorders may occur occasionally with long-term administration.

Special Population Medication

Precautions for children: The dosage for children under 2 years old has not been determined. Precautions for pregnancy and lactation: The drug can pass through the placenta. Although animal experiments have confirmed that it has no effect on the embryo, pregnant women still need to weigh the pros and cons of using the drug. The concentration of the drug in breast milk is 0.6 to 4.1 times the blood concentration. Although no abnormalities have been found in infants, breastfeeding women should use it with caution. Precautions for the elderly: Due to the decline of physiological renal function, the dosage and medication interval of this product need to be adjusted.

Drug Interactions

1. Combined use with zidovudine can cause nephrotoxicity, manifested as deep lethargy and fatigue. 2. Competitive inhibition of organic acid secretion with probenecid. Combined use with probenecid can slow down the excretion of this product, prolong the half-life, and accumulate the amount of drug in the body.
