This product is white or off-white tablets.
Indication
1. Hypertension. This product can be used alone or in combination with other antihypertensive drugs. 2. Chronic stable angina pectoris and variant angina pectoris. This product can be used alone or in combination with other antianginal drugs.
Usage and Dosage
The usual starting oral dose is 5 mg, once a day, and the maximum dose is no more than 10 mg, once a day. Thin people, those with weak constitutions, elderly patients, or those with impaired liver function should start taking the drug from 2.5 mg, once a day; those who take other antihypertensive drugs in combination should also start taking the drug from this dose. The dosage is adjusted according to individual needs, and the adjustment period should be no less than 7-14 days so that the doctor can fully evaluate the patient's response to the dose. However, under the premise of clinical guarantee, the adjustment speed can be accelerated. The recommended dose for the treatment of angina pectoris is 5-10 mg, and the elderly patients or those with impaired liver function need to reduce the dose.
Adverse Reactions
1. This product is well tolerated within the dose range of 10 mg/day, and most adverse reactions are mild to moderate. 2. The discontinuation rate of this product due to adverse reactions is only 1.5%, which is not significantly different from placebo (about 1%). 3. The most common adverse reactions are headache and edema. Dose-related adverse reactions with an incidence of 1% are as follows: edema, dizziness, hot flashes, and palpitations. Adverse reactions that are not clearly related to dose, but have an incidence of more than 1.0% are as follows: headache, fatigue, nausea, abdominal pain, and drowsiness. Among the above adverse reactions, edema, hot flashes, palpitations, and drowsiness occur more frequently in women than in men. 4. Precautions
1. Patients who are allergic to dihydropyridine drugs or any ingredients in this product are prohibited from using this product. 2. Pregnant and lactating women should use it with caution.
Special Population Medication
Precautions for children: Not clear yet. Precautions for pregnancy and lactation: Use with caution in pregnant women and lactating women. Precautions for the elderly: The peak time of the blood concentration of this product is similar in elderly and young patients. The increase in the area under the curve (AUC) and the extension of the elimination half-life in elderly patients have led to a downward trend in the elimination rate. It has been reported that when receiving similar doses of amlodipine, elderly patients have the same good tolerance as young patients. Therefore, normal doses can be used for elderly patients. However, it is appropriate to start with a smaller dose and then gradually increase the dose.
Drug Interactions
1. Cimetidine, grapefruit juice, acidogens: The pharmacokinetics of this product are not changed when used together. 2. Atorvastatin, digoxin, ethanol: This product does not affect their pharmacokinetics. 3. Sildenafil: A single dose of sildenafil (Viagra) in patients with primary hypertension has no effect on the pharmacokinetics of this product. The two drugs produce an independent antihypertensive effect when used together. 4. Warfarin: This product does not change the prothrombin action time of warfarin. 5. Digoxin, phentoin and warfarin: The combination with this product has no effect on the plasma protein binding rate. 6. Anesthetics: Inhaled hydrocarbons combined with this product can cause hypotension. 7. Nonsteroidal anti-inflammatory drugs: Indomethacin in particular can weaken the antihypertensive effect of this product. 8. β-blockers: The combination with this product is well tolerated, but it can cause excessive hypotension and rarely aggravate heart failure.
