    Description

    Tigecycline is a glycylcycline antibiotic developed by Francis Tally and marketed by Wyeth under the brand name Tygacil. It was given a U.S. Food and Drug Administration (FDA) fast-track approval and was approved on June 17, 2005. It was developed in response to the growing prevalence of antibiotic resistance in bacteria such as Staphylococcus aureus and Acinetobacter baumannii. The New Delhi metallo-β-Lactamase multidrug-resistant Enterobacteriaceae has also shown susceptibility to tigecycline.

    Tigecycline is a broad - spectrum antibiotic.

    1. Chemical structure and classification

    Tigecycline belongs to the glycylcycline class of antibiotics. It is a derivative of minocycline. Structurally, it has modifications that give it enhanced antibacterial properties compared to traditional tetracyclines.

    2. Mechanism of action

    It inhibits bacterial protein synthesis. Tigecycline binds to the 30S ribosomal subunit of bacteria, preventing the binding of aminoacyl - tRNA to the A - site of the ribosome. This interference blocks the elongation step of protein synthesis. As a result, bacteria are unable to produce essential proteins needed for their growth, replication, and survival. This mechanism of action is similar to that of other tetracycline - based antibiotics, but tigecycline's binding affinity and antibacterial spectrum are broader in some respects.
