Description	Aripiprazole is an atypical antipsychotic. It is recommended and primarily used in the treatment of schizophrenia and bipolar disorder. Other uses include as an add-on treatment in major depressive disorder, tic disorders, and irritability associated with autism. Aripiprazole is a quinoline derivate and atypical anti-psychotic agent. Aripiprazole has partial agonistic activity at dopamine D2 receptors and serotonin 5-HT1A receptors, as well as potent antagonistic activity on serotonin 5-HT2A receptors.

Aripiprazole, commonly known by the brand name Abilify, is an atypical antipsychotic medication. It is used to treat various mental health conditions. Aripiprazole modulates neurotransmission overactivity in the dopaminergic mesolimbic pathway, which is thought to cause positive symptoms of schizophrenia.

Mechanism of Action

Aripiprazole's mechanism of action is complex and involves several receptor systems:
Dopamine D2 receptors: Aripiprazole functions as a partial agonist at dopamine D2 receptors, modulating neurotransmission in dopaminergic pathways. It can both stimulate and inhibit dopamine at these receptors. This results in reduced positive symptoms and extrapyramidal motor side effects.
Serotonin 5-HT1A receptors: Aripiprazole is also a partial agonist of the postsynaptic serotonin 5-HT1A receptor.
Serotonin 5-HT2A receptors: Aripiprazole acts as an antagonist at the serotonin 5-HT2A receptor. It is a very weak partial agonist of this receptor.
Serotonin 5-HT2B receptors: Aripiprazole has great binding affinity and acts as a potent inverse agonist at the 5-HT2B receptor.
Serotonin 5-HT2C receptors: Aripiprazole is a high-efficacy partial agonist of the postsynaptic 5-HT2C receptor, which may underlie the minimal weight gain seen with this medication.
Serotonin 5-HT7 receptors: Aripiprazole is a very weak partial agonist and functional antagonist of this receptor.
Aripiprazole's actions differ markedly across receptor systems, sometimes acting as an antagonist, sometimes as an inverse agonist, and sometimes as a partial or full agonist. This mixture of actions is consistent with the hypothesis that aripiprazole has "functionally selective" actions.

Applications

Aripiprazole is used to treat a variety of mental health conditions. The desired outcome of antipsychotic agents like aripiprazole in schizophrenia is to inhibit dopaminergic transmission in the limbic system and enhance dopaminergic transmission in the prefrontal cortex.
