Mechanism:A cell-permeable, synthetic tetrapeptide (D-Arg-Dmt-Lys-Phe-NH₂) that selectively concentrates in the inner mitochondrial membrane and reversibly binds to cardiolipin, a phospholipid essential for electron transport chain (ETC) function. By stabilizing cardiolipin, SS-31 restores mitochondrial bioenergetics, reduces cytochrome c peroxidase activity, and prevents oxidative damage in cells with impaired mitochondrial function.
Activity note:SS-31 does not act via traditional receptor agonism. It binds cardiolipin with high selectivity in the inner mitochondrial membrane. Functional EC₅₀ values are context-dependent (cell type, stress model). Key electrophysiology studies report mitochondrial membrane potential restoration at nanomolar concentrations.
