To the suspension of 3-methoxy-4-nitrobenzonitrile (11.4 g, 64 mmol) in ethyl acetate (60 mL) was added 10percent Pd/C (1 g).Step c: To the suspension of 3-methoxy-4-nitrobenzonitrile (11.4 g, 64 mmol) in ethyl acetate (60 mL) was added 10percent Pd/C (1 g). The reaction mixture was vigorously shaken in a Parr under an atmosphere of hydrogen (50 psi) at room temperature for 45 min. After 24 h the reaction was filtered, concentrated, and chromatographed (ethyl acetate/hexanes, 25/75) to yield 1.3 g (60%) of 4-amino-3-methoxybenzonitrile: 1 H NMR (DMSO-d6): delta 7.1 (m, 2H), 6.65 (d, 1H), 5.77 (br s, 2H), 3.79 (s, General procedure: To an ice-cold suspension of nicotinoyl chloride hydrochloride(I) (1.78 g, 10 mmol) and appropriate 4-aminobenzonitrile (IIa-g) (10 mmol, 1.0 equiv) in acetonitrile(50 ml) under nitrogen was added triethylamine (8.1 g, 11.15 ml, 40 mmol, 4.0 equiv) dropwise over 30 min. The resulting suspensionwas stirred at 0 C for 1 h then at room temperatureovernight. The reaction mixture was evaporated, diluted withEtOAc, washed with 10% HCl, brine, dried over anhydrousNa2SO4 and concentrated in vacuo. The resulting solid waswashed with diethyl ether and dried to yield the desired compounds(IIIa-g) in 50-90% yield.
