In a recent development, the European Food Safety Authority (EFSA) has released its evaluation on the safety of a specific β-galactosidase enzyme derived from the non-genetically modified Papiliotrema terrestris strain AE-BLC. This food enzyme, produced by Amano Enzyme Inc., aims to facilitate the production of galacto-oligosaccharides (GOS) from lactose.


After a meticulous assessment, a panel of experts concluded that, under expected usage conditions, the risk of allergenic reactions from dietary exposure to this food enzyme is minimal, though not entirely ruled out. The provided data supported the conclusion that the enzyme does not pose any safety concerns when used as intended.

 

The food enzyme β-galactosidase, also known as β-d-galactoside galactohydrolase (EC 3.2.1.23), was confirmed to be free from viable cells of the production organism, ensuring its purity. Dietary exposure to the food enzyme, measured as total organic solids (TOS), was estimated to be up to 0.441 mg TOS/kg body weight (bw) per day for European populations.

 

Comprehensive genotoxicity tests conducted on the enzyme did not raise any safety concerns. Furthermore, a 90-day oral toxicity study in rats revealed a no observed adverse effect level of 1800 mg TOS/kg bw per day, the highest dose tested. Comparing this level with the estimated dietary exposure, a significant margin of safety, with a minimum margin of exposure of 4082, was demonstrated.

 

To mitigate potential allergic reactions, an examination was conducted to determine if the amino acid sequence of the food enzyme resembled known allergens. Fortunately, no matches were found, further decreasing the likelihood of allergic responses upon dietary exposure.

 

The evaluation conducted by the EFSA underscores the importance of ensuring the safety of enzymes used in food production. With the approval and endorsement of this β-galactosidase enzyme, the industry can confidently explore its applications in the modification of lactose, opening up new avenues for the production of galacto-oligosaccharides (GOS).
