The pharmaceutical segment alone generated $53.6 billion in revenue, showcasing a 3% year-on-year increase. The Rise of the K Drug and HPV Vaccine:

Merck's star performer, the PD-1 monoclonal antibody K drug (Keytruda), continued its upward trajectory in 2023, contributing $25 billion in revenue, a remarkable 21% YoY growth. This phenomenal achievement accounted for approximately 46% of Merck's total drug revenue. Keytruda's sales surpassed the annual sales figures of Comirnaty, the jointly developed vaccine by Pfizer and BioNTech, making it the undisputed "new drug king." Comirnaty held the top spot in the best-selling drug list in 2021 and 2022, with sales figures of $55.9 billion and $55.1 billion, respectively. Pfizer's 2023 annual report revealed global revenue of $11.2 billion for Comirnaty, while BioNTech has yet to disclose its full-year performance. Another standout performer was Merck's HPV nine-valent vaccine, Gardasil/Gardasil9, which achieved an impressive $8.9 billion in sales, representing a 27% YoY growth. The strong demand in the Chinese market, where Gardasil enjoys a monopoly position, played a significant role in its success.

 

Notable Performances and Future Prospects:

Merck's other products in the oncology field also demonstrated commendable growth in 2023. Drugs such as Welireg (HIF-2α inhibitor), Reblozyl (luspatercept), Prevymis (letermovir) for the treatment of cytomegalovirus infection, and the 15-valent pneumococcal vaccine Vaxneuvance (V114) all achieved double-digit or higher growth rates, contributing substantial incremental revenue. The varicella vaccine series also continued to thrive, generating an annual revenue of $2.368 billion, a 4% YoY increase. Additionally, the oral rotavirus vaccine Rotateq, the 15-valent pneumococcal vaccine Vaxneuvance, the 23-valent pneumococcal vaccine, and the hepatitis A vaccine Vaqta garnered impressive sales figures of $769 million, $665 million, $412 million, and $120 million, respectively. Bridion, an anesthesia drug used for reversing neuromuscular blockade, achieved $1.842 billion in revenue, representing a 9% YoY growth.


Future Potential of the K Drug:

Keytruda has been instrumental in driving Merck's performance in recent years and shows no signs of slowing down. Merck continues to invest heavily in developing and promoting Keytruda, which has exhibited remarkable efficacy across more than 40 approved indications, making it the gold standard in several major therapeutic areas. In January 2023, Keytruda received FDA approval for a new indication, allowing its use in combination with chemoradiotherapy for the treatment of stage III-IVA cervical cancer patients. This marked the 39th FDA-approved indication for Keytruda in the United States and the third indication for cervical cancer treatment. Furthermore, Keytruda's expansion in China has been substantial. On December 18, 2023, Merck announced that Keytruda had obtained approval from the National Medical Products Administration (NMPA) in China for first-line treatment in locally advanced unresectable or metastatic HER2-negative gastric or gastroesophageal junction adenocarcinoma when combined with fluoropyrimidine- and platinum-based chemotherapy. This approval signifies Keytruda's 12th indication in China, including its third indication for digestive tract tumors.
