This remarkable feat was largely fueled by the commercial triumph of CARVYKTI®, the company's flagship product. With such an impressive performance in its inaugural year, CARVYKTI® is on track to join the exclusive club of billion-dollar blockbusters.
The Unleashed Potential of CARVYKTI®:
CARVYKTI® is an innovative BCMA CAR-T therapy known as Cilta-cel. It utilizes genetically modified T cells equipped with a chimeric antigen receptor (CAR) to target and eliminate cells expressing BCMA. The CAR protein of CARVYKTI® exhibits high affinity for BCMA-expressing cells, enabling T-cell activation, expansion, and subsequent eradication of the target cells. Clinical data from the CARTITUDE-4 study presented at the 2023 American Society of Hematology (ASH) Annual Meeting demonstrated significant improvements in health-related quality of life for treated patients, as well as a reduction in multiple myeloma burden.
Expanding Commercialization Efforts:
Legendary Biotech, in collaboration with Johnson & Johnson, has obtained conditional marketing approvals for CARVYKTI® in the United States, Japan, and Europe. CARVYKTI® has already been launched in Germany and Austria and has been administered to over 2,500 patients in more than 80 authorized treatment centers worldwide. The commercial demand for CARVYKTI® continues to surge, with the product targeting a sales peak of $5 billion in its first approved year. To meet this growing demand, the companies have been investing heavily in expanding their CAR-T production capacity.
Overcoming Production Challenges:
Like many CAR-T therapies, production capacity has been a significant hurdle for Legendary Biotech and its partner, Johnson & Johnson. The companies have been working diligently to ramp up their CAR-T manufacturing capabilities. Investments have been made to expand the New Jersey facility, the primary site for CARVYKTI® commercial products. Additionally, a second factory in Belgium has commenced production, while a third facility in the Netherlands is currently under construction to support CARVYKTI®'s lentiviral vector production. The companies aim to achieve an annual production capacity of 10,000 doses by the end of 2025.
Future Prospects and Regulatory Expansion:

They have submitted applications to the European Medicines Agency (EMA) and the U.S. Food and Drug Administration (FDA) to broaden CARVYKTI®'s indications. The FDA's Prescription Drug User Fee Act (PDUFA) goal date for the expanded indication review is set for April 5, 2024. Furthermore, positive opinions from the EMA Committee for Medicinal Products for Human Use (CHMP) have been received for CARVYKTI®'s use in the second-line treatment of relapsed or refractory multiple myeloma patients.
