Analysis Of Hot Topics In Pharmacy Research In China
Pharmacy is an important branch ofpharmaceutical science and plays an important and special role in thedevelopment of pharmaceutical science and pharmaceutical industry in China's new situation. The hot topics in pharmacy research in China mainly includetargeted preparations, controlled release preparations, transdermal absorptionpreparations, and drug stability. With the development of science andtechnology, pharmacy has entered a new stage in new materials, newtechnologies, new methods and basic research. This paper reviews and reflectsthe hot topics in pharmacy research by analyzing the basic ideas and hot topicsin pharmacy research in China.
Research on targeted agents
Targeted drug delivery systems candeliver drugs to target organs, reduce the distribution of drugs in normaltissues, improve efficacy, reduce drug dosage, and reduce toxic and sideeffects.
1. Passive targeting agents
The mechanism of passive targetingis: the reticuloendothelial system (RES) has abundant phagocytes, which cantake particles of a certain size (0.1-3um) as foreign bodies in the liver andspleen; larger particles (7-30uln) They cannot be filtered through thecapillary bed and are mechanically trapped in the lungs; however, particlessmaller than 50 nm can enter the bone marrow through the capillary extremities.
2. Active targeting agents
Breast cancer is one of the mostcommon cancers in humans. Some researchers encapsulated anthracyclineanticancer drugs in albumin NP, and then cross-linked anti-human breast cancermonoclonal antibody on the surface of NP, so that NP is like a drug library.When it reaches the cancerous site, the drug can be released by physicaldiffusion to exert its curative effect.
3. Physical and chemical targetingagents
Heat-sensitive Ij and ph-sensitive15, while able to release encapsulated drugs in the target-specificenvironment, cannot deliver drugs to the target in a directed manner.Thermo-sensitive magnetic Ij is magnetic, and the encapsulated drug can bedirected to the target under the control of an external magnetic field.Similarly, this 15 is prepared with thermo-sensitive lipid material. Under theaction of an external heating device in the lesion area, the fluidity of theLIPID membrane of Ij increases, and the encapsulated drug is releasedquantitatively.
Research on controlled releaseformulations
The ideal drug delivery system(DDS) should feature timed, quantitative and targeted drug delivery. Targetedagents can solve the problem of targeted drug delivery, while timed andquantitative drug delivery needs to be realized by controlled release agents.Controlled release agents are mainly divided into self-tuning controlledrelease drug delivery system and pulse release drug delivery system.
1. Self-adjusting drug deliverysystem
The self-regulating drug deliverysystem is a new type of controlled-release drug delivery system thatautomatically adjusts the drug release rate according to needs through aninformation feedback mechanism without external intervention. Because the onsetof the disease changes periodically, DDS is required to actively adjust thedrug release rate according to the needs of the disease. There are severalcontrolled release mechanisms: pH-sensitive polymers; enzyme-substratereactions; pH-sensitive drug solubility; competitive binding; hydrolysisdependent on metal concentration, and more. Some researchers used pH-sensitivematerials to prepare pH-sensitive liposome vesicles, and used the phenomenon ofabnormal pH acidification in tumor, inflammation and infection areas tospecifically release the contained drugs.
2. Pulse drug release system
Pulsed drug delivery systems(PRDDS) deliver drugs rapidly and regularly, both injectable and oral.Researchers have designed a single-dose, multivalent vaccine, in which a singleinjection can produce multiple doses. The basic idea is to make microspheres ofvarying thickness that contain the vaccine and can be released at differenttimes. At the same time, according to the characteristics of cardiovascularsystem diseases such as early morning attacks, oral PRDDS were prepared bycontrolling the types of tablet coating materials or coating thickness andother methods. If taking medicine before bed, a dose can be released in theearly morning pulse to achieve the effect of disease prevention and treatment.
Research on transdermal drugdelivery system
Transdermal therapeutic systems(TTS) are a special case of controlled release formulations. TTS can producelasting, constant and controllable blood drug concentration, reduce toxic andside effects; avoid first-pass effect, improve bioavailability; reduce the painof injection medication; convenient medication and withdrawal; reduce thenumber and dose of administration. The means of promoting the transdermalabsorption of drugs include pharmaceutical, chemical and physical methods.
1. Researchon the effect of pharmacy to promote penetration
The most common method is to usevarious types of percutaneous absorption enhancers (PE). In addition, NP, 15 oremulsion can also be used as a drug carrier to promote drug absorption orcontrol drug release.
2. Research on chemical permeationpromotion
In terms of promoting thetransdermal absorption of drugs, the most commonly used chemical method is tosynthesize prodrugs, so that they have appropriate physical and chemicalproperties such as water-oil partition coefficient, so as to generate a largerpenetration rate during transdermal absorption.
3. Research on the role of physicalpenetration
Physical methods includeiontophoresis, electroporation, and osmotic techniques such as ultrasound,laser, and exotherm.
