Function and Efficacy
Almost all of it is converted into 6-mercaptopurine in the body to work. Because of its slow conversion process, it works slowly. It can inhibit Friend leukemia, inhibit virus infection in mice, inhibit spleen enlargement, and reduce the virus titer in the spleen and plasma. Rats injected with this product intraperitoneally for 4 to 5 months have weight loss, severe anemia and increased reticulocytes. Rabbits given this drug in early pregnancy can cause teratogenesis, mainly affecting limb development. It can have an immunosuppressive effect by interfering with RNA metabolism. If a small dose exists in the culture medium for a long time, it can inhibit the killing cell effect of sensitized lymphocytes in vitro.

Appearance
This product is light yellow tablet.
Indication
1. Acute and chronic leukemia, with good short-term efficacy for chronic myeloid leukemia, rapid effect, but short remission period; 2. Acquired hemolytic anemia, idiopathic thrombocytopenic purpura, systemic lupus erythematosus; 3. Chronic rheumatoid arthritis, chronic active hepatitis (hepatitis related to autoimmunity), primary biliary cirrhosis; 4. Hyperthyroidism, myasthenia gravis; 5. Others: chronic nonspecific ulcerative colitis, segmental enteritis, polyradiculitis, lupus nephritis, proliferative nephritis, Wegener's granulomatosis, etc.
Usage and Dosage
1. Oral administration: 1.5 mg to 4 mg/kg per day, once a day or divided into several times; 2. For allogeneic transplantation, 2 mg to 5 mg/kg per day, once a day or divided into several times; 3. For leukemia, 1.5 mg to 3 mg/kg per day, once a day or divided into several times.
Adverse Reactions
It is similar to mercaptopurine but slightly less toxic. It can cause bone marrow suppression, liver damage, teratogenesis, skin rash, and occasionally muscle atrophy.
Precautions
Patients with known hypersensitivity to this product are contraindicated.

Drug Interactions
Allopurinol can inhibit the metabolism of mercaptopurine (the latter is the active metabolite of thiopurine) into inactive products, resulting in increased toxicity of mercaptopurine. When the two must be taken at the same time, the dose of thiopurine should be greatly reduced. Thiopurine can reduce the inactivation rate of 6-mercaptopurine, which is inactivated by the following ways: S-methylation of the enzyme, oxidation unrelated to the enzyme, or conversion to thiourate by xanthine oxidase, etc. Thiopurine can react with sulfhydryl compounds such as glutathione, slowly releasing 6-mercaptopurine in tissues and acting as a prodrug.
