    1. Description

    5-Amino-1-methylquinolinium iodide is a selective nicotinamide N-methyltransferase inhibitor that targets the substrate-binding site. Its chemical structure is a quaternary ammonium salt formed by introducing an amino group at the 5-position of 1-methylquinoline. It appears as a light red to dark brown powder with a purity of over 98% and is soluble in dimethyl sulfoxide (DMSO). Storage conditions require keeping the product sealed and protected from light at -20°C. This compound exerts its effect by selectively inhibiting NNMT activity and has no significant impact on related methyltransferases or enzymes in the NAD⁺ salvage pathway.

    2. Application

    (1) Research on metabolic diseases: Used in preclinical studies of metabolic disorders such as obesity and type 2 diabetes; subcutaneous administration in high-fat diet-induced obese mouse models has demonstrated therapeutic effects.

    (2) Regenerative Medicine Research: Used in studies on muscle injury repair; subcutaneous administration in muscle injury models promotes myocyte differentiation and regeneration.

    (3) Oncology Research: Used in mechanism studies of tumors such as ovarian cancer; it has demonstrated antitumor activity in peritoneal metastasis models.

    (4) Pharmacokinetic Research: Used as a tool compound for NNMT inhibition to study its absorption, distribution, metabolism, and excretion characteristics in vivo.

    3. Main Efficacy

    (1) Selective inhibition of NNMT activity: By targeting the NNMT substrate-binding site, this compound selectively inhibits the enzyme’s methylation activity without affecting the function of related methyltransferases or enzymes in the NAD⁺ salvage pathway. In adipocytes, it reduces the levels of 1-methylnicotinamide, a product of the NNMT reaction, in a dose-dependent manner.

    (2) Promotion of myoblast differentiation and muscle regeneration: Treatment in myoblasts significantly increases the degree of myocyte differentiation. By inhibiting NNMT activity, it regulates intracellular NAD⁺ levels and methylation metabolism, thereby promoting muscle stem cell activation and myofiber regeneration.

    (3) Anti-adipogenic effects: Treatment in preadipocytes reduces lipid droplet accumulation and downregulates the expression of adipogenesis marker genes. By influencing NNMT activity in adipocytes, it regulates energy metabolism and lipid synthesis.

    (4) Oral bioavailability: Pharmacokinetic studies indicate that the compound exhibits good plasma exposure following oral administration, suggesting favorable oral absorption characteristics.
