1. Description

In humans, ecdysone promotes protein synthesis, helps eliminate cholesterol from the body, lowers blood lipid levels, and inhibits blood sugar spikes. It is traditionally used to treat rheumatoid arthritis. In sericulture, it is used to shorten the life cycle of silkworms and promote silk production and cocoon formation; in aquaculture, it is widely used in the farming of shrimp and crabs; in cosmetics, ecdysone is also used as a special additive; when added to cosmetics, ecdysone can prevent skin dryness and make the skin smoother and more delicate. Ecdysone influences all developmental stages of insects, from hatching larvae to adults; consequently, it is used to control or eliminate agricultural pests and is currently being developed and applied as a third-generation pesticide. Ecdysone also exerts certain effects on the physiological processes of crops, offering promising prospects for development and application as a crop yield-enhancing agent.

2. Application

In humans, ecdysone promotes collagen synthesis, helps prevent arrhythmia, and combats fatigue; it promotes cell growth, stimulates dermal cell division, and helps eliminate cholesterol from the body; it lowers blood lipid levels and inhibits blood sugar spikes; it also has physiological activities such as unblocking meridians, dispelling dampness, and relieving pain; it is a natural anticancer agent; since its introduction in 1976, ecdysone from Dew Grass has been formulated into tablets and ointments. Traditionally used to treat rheumatoid arthritis, it has shown good efficacy in patients with various symptoms of rheumatism and arthritis. Its active ingredient, 20-hydroxyecdysterone, has been clinically proven to directly lower blood glucose levels, with an overall efficacy rate of 84% for Type 2 diabetes, and is particularly effective in alleviating the “three excesses and one deficiency” symptoms commonly experienced by diabetic patients.

3. Main Efficacy

(1) Promotion of muscle protein synthesis and hypertrophy: In C2C12 myoblasts, 20-hydroxyecdysterone increases protein synthesis by up to 20% by activating the PI3K/Akt signaling pathway and elevating cytoplasmic Ca²⁺ levels. This effect can be blocked by GPCR inhibitors and selective estrogen receptor β antagonists, suggesting a membrane receptor-mediated mechanism. The expression of myostatin, a negative regulator of muscle growth, decreases following 20E treatment.

(2) Activation of the MAS1 receptor: Recent studies have identified the activation of the MAS1 receptor (a key component of the renin-angiotensin system) as a primary mechanism of action for 20-hydroxyecdysone, which may explain its multifunctional effects observed in various animal models. Angiotensin I-7 (an endogenous ligand of the MAS receptor) exhibits effects similar to 20E in reducing myostatin and promoting myoblast proliferation and myotube formation; these effects can be reversed by MAS receptor-specific antagonists and siRNA silencing.

(3) Antioxidant and neuroprotective effects: Protects PC12 cells from CoCl₂-induced cellular damage by inhibiting reactive oxygen species (ROS) production, reducing mitochondrial membrane depolarization, inhibiting cytochrome c release, regulating the Bax/Bcl-2 ratio, and inhibiting caspase-3 activation.
